Microbiome composition indicate dysbiosis and lower richness in tumor breast tissues compared to healthy adjacent paired tissue, within the same women

Maria Valeria Esposito et al.

Summary: Breast cancer is the most common malignancy in women and the imbalance of gut microbiota has been associated with its development. This study analyzed the microbiome of breast tissue in 34 women with breast cancer and found significant differences in bacterial composition and richness between healthy and tumor tissues. These differences may have diagnostic and therapeutic implications, but further research is needed to determine if they are merely an association or a causal effect in breast cancer.

Comprehensive metabolomics expands precision medicine for triple-negative breast cancer

Yi Xiao et al.

Summary: Metabolic reprogramming in triple-negative breast cancer (TNBC) was systematically characterized in this study, resulting in the construction of a large metabolomic atlas. The study classified TNBCs into three distinct metabolomic subgroups and identified subtype-specific metabolites as potential therapeutic targets. The findings suggest the clinical significance of TNBC metabolomics and provide valuable resources for precision treatment of TNBC.

The Kynurenine Pathway and Cancer: Why Keep It Simple When You Can Make It Complicated

Roumaissa Gouasmi et al.

Summary: The kynurenine pathway plays important roles in tumor development by protecting organs from immune reactions and providing essential metabolites to cells. Abnormal activation of this pathway promotes tumor growth, metastasis, and chemoresistance. The complex regulation of this pathway poses challenges for clinical interventions.

Circulating Carnitine Levels and Breast Cancer: A Matched Retrospective Case-Control Study

Jiayi Zhang et al.

Summary: This study found an association between blood carnitine compounds and breast cancer through a retrospective case-control study. Higher levels of malonylcarnitine, decenoylcarnitine, and decadienoylcarnitine were associated with decreased odds of breast cancer, while higher levels of butyrylcarnitine were associated with increased odds of the disease.

Metabolic-Pathway-Based Subtyping of Triple-Negative Breast Cancer Reveals Potential Therapeutic Targets

Yue Gong et al.

Summary: This study investigated metabolic dysregulation in TNBCs using a multi-omics database, identifying three heterogeneous metabolic-pathway-based subtypes with distinct metabolic features. These subtypes had different prognoses, molecular subtype distributions, and genomic alterations, as well as varying sensitivities to metabolic inhibitors targeting fatty acid synthesis and glycolysis. Additionally, inhibition of lactate dehydrogenase could enhance tumor response to anti-PD-1 immunotherapy in specific TNBC subtypes.

Integrated Molecular Characterization to Reveal the Association between Kynurenine 3-Monooxygenase Expression and Tumorigenesis in Human Breast Cancers

Yuk-Wah Tsang et al.

Summary: The study revealed that Kynurenine 3-monooxygenase (KMO) is highly expressed in various tumors, particularly in invasive ductal breast carcinoma, and positively correlated with malignant clinical features in breast cancer patients. The top ten KMO-correlated genes are chemokines and pro-inflammatory cytokines known to be involved in cancer progression, indicating that KMO may facilitate breast cancers by regulating inflammatory responses synergistically with these hub genes. These findings highlight the tumor-promoting role of KMO in breast cancers and suggest its potential as a biomarker for prognosis prediction.

Characterizing the breast cancer lipidome and its interaction with the tissue microbiota

Natasa Giallourou et al.

Summary: This study identified unique lipid signatures in breast tissue of women with breast cancer compared to healthy individuals, and observed correlations between specific bacterial groups and certain lipids in the tissue. Further investigation into the cause-effect relationships and contributions to disease patho-mechanisms is needed for this globally impacting disease.

Relationship of established risk factors with breast cancer subtypes

Anne Marie McCarthy et al.

Summary: Breast cancer is a heterogeneous disease with different subtypes based on ER, PR, and HER2 expression. This study found that Black women had 2.7 times higher risk of TNBC compared to white women, breast density and BMI were associated with different subtypes, and TNBC was more likely to be interval cancer.

Glucose Metabolism and Glucose Transporters in Breast Cancer

Eunah Shin et al.

Summary: Breast cancer, the most common malignancy in women worldwide, is closely associated with glucose metabolism, with high expressions of glucose metabolism-related enzymes and transporters. The tumor microenvironment of breast cancer also involves various stromal cells, contributing to the heterogeneous metabolic status of the disease.
