Towards controlled drug delivery in brain tumors with microbubble-enhanced focused ultrasound

Scott Schoen et al.

Summary: Brain tumors located in the central nervous system (CNS) present a unique challenge due to limited drug access to the brain tumor microenvironment (TME). Recent advancements in microbubble ultrasound contrast agents and ultrasound energy have shown promise in enhancing drug delivery to brain tumors. This technology has the potential to improve the effectiveness of both traditional chemotherapy and novel immunotherapeutic agents. Early clinical studies have demonstrated safety, and ongoing Phase I/II trials are testing the preclinical findings.

Engineering Macrophage Exosome Disguised Biodegradable Nanoplatform for Enhanced Sonodynamic Therapy of Glioblastoma

Tingting Wu et al.

Summary: Sonodynamic therapy (SDT) is a promising cancer treatment modality for glioblastoma (GBM) due to its high tissue penetration and minimal damage to normal tissues. However, the blood-brain barrier (BBB) and hypoxic microenvironment present challenges to SDT efficiency. In this study, a biodegradable nanoplatform (CSI@Ex-A) was developed, which could effectively penetrate the BBB and target cancer cells. The nanoplatform releases hydrogen peroxide (H2O2) and oxygen (O-2) to relieve tumor hypoxia, thereby enhancing the efficacy of SDT.

Contribution of Oxidative Stress Induced by Sonodynamic Therapy to the Calcium Homeostasis Imbalance Enhances Macrophage Infiltration in Glioma Cells

Lei Chen et al.

Summary: Sonodynamic therapy (SDT) is a non-invasive technique that destroys tumor cells through mitochondrial respiratory chain damage and increased intracellular reactive oxygen species (ROS) and calcium overload levels. This study found that the early application of the mechanosensitive Ca2+ channel antagonist GsMTx4 can enhance the anti-tumor effect of SDT by promoting the dissociation and polymerization of the Ca2+ lipid complex and increasing oxidative stress levels. The study also revealed the mechanism of intracellular Ca2+ signal regulation, lipid metabolism, and recruitment of macrophages in glioma treatment.
