NK cells-directed therapies target circulating tumor cells and metastasis

Hassan Dianat-Moghadam et al.

Summary: Metastasis is a major cause of cancer-related deaths, with CTCs playing a critical role in this process. Understanding the interaction between immune cells and CTCs, particularly the role of NK cells in recognizing and eliminating CTCs, may lead to new targeted immunotherapies for metastatic diseases.

LncRNA MEG3 suppresses PI3K/AKT/mTOR signalling pathway to enhance autophagy and inhibit inflammation in TNF-α-treated keratinocytes and psoriatic mice

Zhan-Li Tang et al.

Summary: The study reveals that downregulation of lncRNA MEG3 is associated with enhanced inflammation and suppressed autophagy in psoriasis, while overexpression of lncRNA MEG3 can reverse these effects, potentially through the PI3K/AKT/mTOR signaling pathway. Therefore, lncRNA MEG3 may serve as a therapeutic target for psoriasis.

Landscape of innate lymphoid cells in human head and neck cancer reveals divergent NK cell states in the tumor microenvironment

Uriel Y. Moreno-Nieves et al.

Summary: Despite the reported antitumor functions of NK cells, their contribution to tumor control in humans remains controversial due to incomplete understanding of the NK cell states within the tumor microenvironment (TME). This study demonstrates that peripheral circulating NK cells differentiate into two divergent pathways within the TME, resulting in different end states, with one having potent in vivo antitumor activity. This finding provides insight into the origin of certain NK cell states within the TME and identifies specific NK cell phenotypes with the greatest antitumor activity.

Circulating and Tumor-Infiltrating NK Cells From Clear Cell Renal Cell Carcinoma Patients Exhibit a Predominantly Inhibitory Phenotype Characterized by Overexpression of CD85j, CD45, CD48 and PD-1

Andrea Ziblat et al.

Summary: The study found that NK cells from ccRCC patients exhibit different activation characteristics compared to those from healthy donors, with NK cells in the tumor microenvironment showing higher activation markers and a more inhibitory phenotype. Overexpression of specific molecules is associated with an NK cell infiltration signature, potentially serving as new biomarkers for ccRCC patients and providing novel targets for immunotherapy.
