A Metabolomic Aging Clock Using Human Cerebrospinal Fluid

Summary: Quantifying the physiology of aging is crucial for understanding age-related diseases and healthy aging heterogeneity. Recent studies indicate that omic clocks provide measures of biological age, with metabolomic profiles able to predict chronological age within 10 years in healthy subjects. Predictive models for age using metabolomic profiles show increased prediction error in subjects with Alzheimer's and Parkinson's disease, possibly indicating differences in the relationship between the metabolome and chronological age in these diseases.

The levels of nicotinamide adenine dinucleotide (NAD(+)) were found to be significantly lower in older adults, and this decrease was more pronounced in physically impaired individuals. However, exercise-trained older individuals had NAD(+) levels similar to those of younger individuals, indicating a positive effect of physical activity on NAD(+) abundance and muscle functioning.
