Synthesis and antitumor activity of new tetrahydrocurcumin derivatives via click reaction

Meitao Duan et al.

Summary: Three new derivatives of tetrahydrocurcumin were synthesized as potent antitumor agents using 'click chemistry' catalyzed by copper(II), with compound 6 showing significant inhibitory activity against HCT-116 cell line. Further modifications of tetrahydrocurcumin structure are suggested to improve its anticancer activity.

An Overview of the Biological Evaluation of Selected Nitrogen-Containing Heterocycle Medicinal Chemistry Compounds

Oluwakemi Ebenezer et al.

Summary: Heterocyclic compounds are natural compounds with favorable properties and significant pharmaceutical importance. They have diverse applications as smart biomimetics and possess active pharmacophores in complex structures, making them indispensable in drug discovery. Among different heterocyclic ring systems, nitrogen heterocyclic compounds are more abundant in nature and have considerable pharmacological significance. This review highlights pioneering studies in the biological assessment of nitrogen-containing compounds, such as triazoles, tetrazoles, imidazole/benzimidazoles, pyrimidines, and quinolines. It covers publications from April 2020 to February 2022 and will be beneficial for researchers in medicinal chemistry and pharmacology.

Design, synthesis and pharmacological evaluation of tricyclic derivatives as selective RXFP4 agonists

Lin Lin et al.

Summary: Relaxin family peptide receptors RXFPs are potential therapeutic targets for neurological, cardiovascular, and metabolic diseases, with RXFP3 and RXFP4 identified as promising targets for treatment. A new selective RXFP4 agonist was discovered through high-throughput screening, with structural modifications leading to a compound with improved selectivity and efficacy for RXFP4 compared to the original scaffold.

Synthesis of Sterically Hindered α-Hydroxycarbonyls through Radical-Radical Coupling

Kenji Ota et al.

Summary: This method synthesizes sterically hindered alpha-hydroxy carbonyl compounds through radical-radical coupling. It allows for the use of primary, secondary, and tertiary aliphatic carboxylic acids to introduce various alkyl substituents onto ketone moieties of alpha-ketocarbonyls under mild reaction conditions.

Diversity-Oriented Synthesis of 1,2,4-Triazols, 1,3,4-Thiadiazols, and 1,3,4-Selenadiazoles from N-Tosylhydrazones

Zeyang Wei et al.

Summary: A diversity-oriented synthesis for 1,2,4-triazols, 1,3,4-thiadiazols, and 1,3,4-selenadiazoles from N-tosylhydrazones was developed, with NH2CN, KOCN, KSCN, and KSeCN as odorless sources. Two different pathways were proposed, and N-tosylhydrazonoyl chlorides were formed in situ in the presence of NCS.

Divergent Synthesis of Substituted Amino-1,2,4-triazole Derivatives

Fenghai Zhao et al.

Summary: An efficient assembly of disubstituted 1,2,4-triazoles was achieved by cyclization of readily accessible N'-nitro-2-hydrocarbylidene-hydrazinecarboximidamides with moderate to excellent yields under mild reaction conditions. This divergent synthetic strategy allows for the formation of amino-1,2,4-triazoles under acidic conditions and nitroimino-1,2,4-triazoles under neutral conditions with water as the solvent.

Deep Eutectic Solvents (DESs) and Their Application in Biosensor Development

Rossella Svigelj et al.

Summary: Deep Eutectic Solvents (DESs) are a new class of solvents characterized by a remarkable decrease in melting point compared to those of the starting components. They have applications in organic synthesis, electrochemistry, and bio-catalysis, showing improved biodegradability and lower toxicity. DESs may have potential use for biosensor development and can solubilize a wide range of molecules for the assembly of new devices.
