Summary: Dopaminergic dysfunction is associated with genetic risk for psychosis, particularly in individuals with 22q11.2 deletion. This study found increased striatal dopamine synthesis capacity in individuals with 22q11.2 deletion compared to healthy controls and individuals with 22q11.2 duplication. Furthermore, dopamine synthesis was positively correlated with the severity of psychosis-risk symptoms.

Summary: Schizophrenia patients show increased striatal dopamine synthesis capacity, possibly due to NMDAR hypofunction and PV neuronal dysfunction. A mouse model mimicking this dopamine pathology was developed, with sub-chronic ketamine treatment increasing dopamine synthesis capacity and locomotor activity. Activation of midbrain dopamine neurons and PV interneurons reversed these effects, indicating potential therapeutic targets.

A potential biomarker for treatment stratification in psychosis: evaluation of an [18F] FDOPA PET imaging approach

Summary: The study evaluated the potential of [F-18]FDOPA PET imaging to identify non-responders to antipsychotic treatment using linear and non-linear machine learning analyses and found good test-retest reproducibility and predictive power in distinguishing responders from non-responders. The economic analysis indicated potential healthcare cost savings by using [F-18]FDOPA PET to fast-track treatment-resistant patients to clozapine. Overall, the findings suggest that [F-18]FDOPA PET dopamine imaging has the potential to serve as a biomarker to guide treatment choice for patients with schizophrenia.

Genetic high-risk individuals also exhibited reduced variability of striatal D2/D3 receptor availability. However, significant heterogeneity between studies suggests the presence of potentially clinically meaningful differences that cannot be ruled out.
