The RASopathies: from pathogenetics to therapeutics

Summary: The genetic disorder CFC3 syndrome, caused by variants in MAP2K1 gene, is mainly characterized by short stature, facial dysmorphism, cardiovascular abnormalities, skin changes, and intellectual disability. A study evaluated the pathological changes in the upper respiratory tract, orthodontic issues, as well as voice, speech and hearing abnormalities in an 11-year-old boy with CFC3 syndrome, revealing issues such as adenoid hypertrophy, narrowing of the nasal passages, laryngeal asymmetry, bilateral sensorineural hearing loss, and speech pathology. This supports the consideration of phoniatric, audiological, orodental, and speech problems as features of CFC3 syndrome type 3 phenotype due to a pathogenic variant in MAP2K1.

Congenital polyvalvular disease expands the cardiac phenotype of the RASopathies

Summary: RASopathies are a group of genetic syndromes with cardiovascular abnormalities, including pulmonary valvular stenosis and hypertrophic cardiomyopathy, while the association of CPVD with RASopathies requires further investigation. Our study found that CPVD was present in 2.5% of individuals in the RASopathy cohort, mainly affecting the aortic and pulmonic valves, with a lower percentage reported in existing literature.
