Summary: Urinary extracellular vesicles (uEV) are considered a potential source of biomarkers for the health and diseases of the urogenital system, but there are challenges in standardizing the collection and analysis process. Storage temperature, time, and format were found to impact the quality of uEV isolates. The transcriptome of uEV could be preserved in urine or isolated uEV even after long-term storage, but storage at -20 degrees C degraded certain components of the transcriptome and EV protein markers.

The research found a strong correlation between urine particle and creatinine concentrations, as well as effects of water loading on uEV size and protein detection. Additionally, using detergent improved intracellular-epitope recognition in uEVs and suggested specific binding of exosome markers CD9 and CD63 with distal nephron markers.
