p53 in ferroptosis regulation: the new weapon for the old guardian

Yanqing Liu et al.

Summary: p53 plays a crucial role in tumor suppression through modulating ferroptosis, a form of iron-dependent non-apoptotic cell death. Through its metabolic targets, p53 can regulate the cellular response to ferroptosis inducers. This review also discusses the potential of targeting p53-mediated ferroptotic responses for the treatment of human cancers and other diseases.

Metformin attenuates hyperlipidaemia-associated vascular calcification through anti-ferroptotic effects

Wen-Qi Ma et al.

Summary: The research showed that Met inhibits hyperlipidaemia-associated calcium deposition in the rat aortic tissue. In vitro, treatment of VSMCs with PA stimulates ferroptosis concomitant with increased calcium deposition in VSMCs, while pretreatment with Met attenuates these effects. This study suggests that targeting POSTN in VSMCs may provide a new strategy for vascular calcification prevention or treatment.

Broadening horizons: the role of ferroptosis in cancer

Xin Chen et al.

Summary: Ferroptosis is an iron-dependent form of regulated cell death driven by excessive lipid peroxidation, and can be induced by various agents to suppress tumor growth while also potentially triggering inflammation-associated immunosuppression. The extent of ferroptosis's impact on tumor biology and its interactions with other signaling pathways are still under investigation.

DHODH-mediated ferroptosis defence is a targetable vulnerability in cancer

Chao Mao et al.

Summary: The study reveals the mechanism of ferroptosis induced by inhibiting GPX4 in cancer cells, as well as the potential pathways to enhance or inhibit ferroptosis by intervening in relevant metabolic pathways. Therapeutic strategies targeting different levels of GPX4 expression could be a promising new approach for cancer treatment.

iPLA2β-mediated lipid detoxification controls p53-driven ferroptosis independent of GPX4

Delin Chen et al.

Summary: The study identifies iPLA2 beta as a critical regulator for p53-driven ferroptosis independent of GPX4. Loss of iPLA2 beta does not have obvious effect on normal cells, but it plays an essential role in regulating ferroptosis upon ROS-induced stress. iPLA2 beta is a promising therapeutic target for activating ferroptosis-mediated tumor suppression without serious toxicity concerns.

Aberrant splicing isoforms detected by full-length transcriptome sequencing as transcripts of potential neoantigens in non-small cell lung cancer

Miho Oka et al.

Summary: This study utilized long-read sequencing to identify novel splicing isoforms in non-small-cell lung cancers and potential neoantigens. The analysis revealed that disruptions in NMD and splicing factors lead to an increase in aberrant transcripts. Evaluation of isoforms binding affinities to HLA molecules indicated the generation of neoantigen candidates. Furthermore, the study found a correlation between disruption of NMD factors and the number of splicing isoforms in lung adenocarcinoma data from TCGA.
