Genetic etiology of non-syndromic hearing loss in Europe

Ignacio del Castillo et al.

Summary: The genetic etiology of non-syndromic hearing impairment (NSHI) in Europe is complex and involves a large number of genes, with variations in mutation frequencies and most common mutations across different populations. DFNB1 is the most common form of autosomal recessive NSHI, while DFNA22 and DFNA8/12 are the most common forms of autosomal dominant NSHI.

The Human Phenotype Ontology in 2021

Sebastian Koehler et al.

Summary: The Human Phenotype Ontology (HPO) serves as a global standard for phenotype exchange, with recent major extensions in neurology, nephrology, immunology, pulmonology, and other fields. Efforts have been made to improve computational definitions of phenotypic abnormalities across HPO and multiple phenotype ontologies for animal disease models, benefiting software accuracy and cross-species phenotype matching. Recent initiatives include translating HPO into indigenous languages and advancing its use in electronic health record systems.

Frequency of carriers for rare recessive Mendelian diseases in a Brazilian cohort of 320 patients

Caio Robledo D'Angioli Costa Quaio et al.

Summary: This study utilized whole exome sequencing to analyze the genetic status of Brazilian patients, revealing a high prevalence of carriers with pathogenic genes for rare genetic diseases. The sensitivity of carrier screening panels was found to be low, and it was estimated that a large proportion of variants would not be detected by these panels. The combined frequencies of autosomal recessive diseases were calculated, providing insights into the clinical and social burden of recessive diseases at the population level.

Whole-Exome Sequencing Reveals a Rare Variant of OTOF Gene Causing Congenital Non-syndromic Hearing Loss Among Large Muslim Families Favoring Consanguinity

Mohd Fareed et al.

Summary: This study aimed to decipher the genetic cause of prelingual NSHL in a large affected Muslim consanguineous families in India using whole-exome sequencing (WES). A rare non-sense homozygous mutation in the OTOF gene was identified, which has not been previously reported in existing databases. In addition to the pathogenic OTOF variant, novel and recurrent mutations in other genes were also observed in this study.
