A Small-Plaque Isolate of the Zika Virus with Envelope Domain III Mutations Affect Viral Entry and Replication in Mammalian but Not Mosquito Cells

Summary: An attenuated Zika virus (ZIKV) isolate from a Thai patient, which was passaged in PDK cells, displayed both big and small plaque-forming viruses. Two small-plaque isolates were selected and characterized as attenuated ZIKV candidates. In vitro experiments showed reduced titers for small-plaque isolates in Vero cells early post-infection, potentially due to diminished entry capabilities. These isolates also exhibited lower neurovirulence in newborn mice. Genomic analysis identified three unique mutations that likely affect virus entry, tropism, and virulence. These findings have important implications for ZIKV vaccine development.
