Summary: The purpose of this trial was to study the feasibility of combined programmed death protein ligand 1/cytotoxic T-lymphocyte-associated protein 4 inhibition with radiotherapy, and to analyze the efficacy of the treatment scheme consisting of induction chemoimmunotherapy followed by chemotherapy-free radioimmunotherapy (RIT) after intratumoral CD8+ immune cell-based patient selection. The trial showed that RIT is feasible and the treatment scheme has promising progression-free survival.

Summary: In the Phase 3, placebo-controlled PACIFIC trial, consolidative durvalumab after chemoradiotherapy showed significant improvements in overall survival and progression-free survival for patients with unresectable, stage III NSCLC, with consistent benefits observed approximately 4 years after treatment. Durvalumab demonstrated durable benefits with a higher 4-year survival rate and progression-free survival rate compared to placebo.

Comparison of DNA repair and radiosensitivity of different blood cell populations

Summary: The research investigated the radiation sensitivity of different human blood cell populations obtained from healthy volunteers. T and B cells were found to be highly sensitive to radiation, but displayed DNA repair competence. In terms of cell death/apoptosis, T cells, NK cells, and B cells showed higher sensitivity compared to monocytes, macrophages, and immature dendritic cells.

Summary: The study identified a predictive LIPS for the survival of cancer patients treated with PD-1/PD-L1 ICI, based on immune cell subsets in peripheral blood. It was found that LIPS showed high accuracy in predicting overall survival and progression-free survival, unaffected by clinicopathological features. NKT cells and neutrophils of the LIPS can serve as dynamic predictive biomarkers for survival outcomes after the first administration of ICI.
