Comprehensive Characterization of RNA-Binding Proteins in Colon Adenocarcinoma Identifies a Novel Prognostic Signature for Predicting Clinical Outcomes and Immunotherapy Responses Based on Machine Learning

Ye Miao et al.

Summary: A multi-gene signature was developed based on the expression profile of RNA-binding proteins (RBPs) to predict prognosis and immunotherapy response in patients with colon adenocarcinoma (COAD). The signature consisted of 8 genes and was able to categorize patients into high- and low-risk populations with different outcomes. The signature outperformed five other prediction models and a nomogram was generated for quantifying overall survival (OS) rate. Additionally, the study found that high-risk populations had enhanced immune evasion capacity, while low-risk populations might benefit from immunotherapy, particularly the combination of PD-1 and CTLA4 inhibitors.

Summary: Accumulating evidence suggests that ferroptosis and pyroptosis play important roles in the tumorigenesis of low-grade glioma (LGG). This research aimed to classify molecular subtypes of LGG and identify a novel multigene signature based on ferroptosis- and pyroptosis-related genes. The study found that the FPRGs had prognostic value and developed a six-gene prognostic signature that outperformed other recognized signatures in predicting the survival probability of LGG patients.

Summary: This study aimed to identify hub genes impacting clinical prognosis for hepatocellular carcinoma (HCC) patients and develop an endoplasmic reticulum (ER)-related prognostic model. Through various analyses, we identified five hub genes associated with ER and categorized HCC patients into high-risk and low-risk groups. Patients in the low-risk group had better overall survival and relapse-free survival. The study also revealed a link between ER-related biological processes and tumor infiltration of immune cells.

Siglec15 shapes a non-inflamed tumor microenvironment and predicts the molecular subtype in bladder cancer

Jiao Hu et al.

Summary: Siglec15 is specifically overexpressed in the tumor microenvironment (TME) of various cancers, likely designing a non-inflamed TME in bladder cancer (BLCA). High expression of Siglec15 in BLCA correlates with lower immune infiltration, lower response to cancer immunotherapy, and a higher incidence of hyperprogression. A combination of anti-Siglec15 and cancer immunotherapy may be a more effective strategy than monotherapy, while an immune risk score (IRS) could accurately predict prognosis and response to treatment.
