Immune suppressive checkpoint interactions in the tumour microenvironment of primary liver cancers

Guoying Zhou et al.

Summary: Liver cancer, particularly hepatocellular carcinoma and cholangiocarcinoma, presents challenges in immune-mediated tumour control due to the immunosuppressive tumour microenvironment. Although key co-inhibitory immune checkpoint interactions play a critical role, only a minority of patients respond to monotherapy using approved checkpoint inhibitor antibodies. Further research is needed to identify alternative immune checkpoints and explore combined targeting of co-inhibitory and co-stimulatory pathways to overcome immunosuppression.

Beyond conventional immune-checkpoint inhibition - novel immunotherapies for renal cell carcinoma

David A. Braun et al.

Summary: The management of advanced-stage renal cell carcinoma has been transformed by the development of immune-checkpoint inhibitors (ICIs). However, most patients do not derive durable clinical benefit from these agents. Innovations in immunotherapy for RCC, including ICIs with novel targets, are now in clinical development.

Construction and characterization of rectal cancer-related lncRNA-mRNA ceRNA network reveals prognostic biomarkers in rectal cancer

Guoying Cai et al.

Summary: Rectal cancer is a significant cause of cancer-related deaths globally. This study identified highly correlated lncRNA-mRNA pairs with strong prognostic effects, constructed a network based on the ceRNA theory, and uncovered key regulators in tumourigenesis through topological analysis and hub subnetwork construction. The findings provide promising clues for biological prognosis of rectal cancer.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Open-Label, Single-Arm Phase II Study of Pembrolizumab Monotherapy as First-Line Therapy in Patients With Advanced Clear Cell Renal Cell Carcinoma

David F. McDermott et al.

Summary: Single-agent pembrolizumab demonstrated promising antitumor activity as a first-line treatment in untreated patients with advanced ccRCC, with durable responses observed across different categories. The safety and tolerability profile of pembrolizumab monotherapy was consistent with what has been observed in other tumor types.

Six distinct NFκB signaling codons convey discrete information to distinguish stimuli and enable appropriate macrophage responses

Adewunmi Adelaja et al.

Summary: The dynamics of NF kappa B signaling can classify immune threats through six signaling codons, and confusion of these codons may underlie the etiology of some inflammatory diseases.NF kappa B activity determines the type of gene expression and response in macrophages. An oscillatory trajectory is a hallmark of responses to cytokine stimuli.

From Bench to Bedside: How the Tumor Microenvironment Is Impacting the Future of Immunotherapy for Renal Cell Carcinoma

Jonathan Anker et al.

Summary: Immunotherapy has revolutionized the treatment of many cancer types, with particular progress seen in the treatment of renal cell carcinoma. While clinical benefit from immunotherapy is limited to certain patients, combination-based approaches have shown improved outcomes. Ongoing translational work has helped illuminate response mechanisms and resistance to immunotherapy, shaping the future of treatment for RCC.

METTL1 promotes hepatocarcinogenesis via m7G tRNA modification-dependent translation control

Zhihang Chen et al.

Summary: The N-7-methylguanosine (m(7)G) modification is a common transfer RNA (tRNA) modification in humans. Elevated levels of METTL1 and WDR4 in hepatocellular carcinoma (HCC) are associated with advanced tumor stages and poor patient survival. Mechanistically, METTL1-mediated tRNA m(7)G modification promotes the translation of target mRNAs with higher frequencies of m(7)G-related codons, providing new insights into the role of misregulated tRNA modifications in liver cancer.

Evolution of Methyltransferase-Like (METTL) Proteins in Metazoa: A Complex Gene Family Involved in Epitranscriptomic Regulation ana Other Epigenetic Processes

Juliet M. Wong et al.

Summary: The study investigates the evolutionary mechanisms of 33 individual METTL proteins in animals, revealing that they are widely distributed in the animal kingdom and mostly originated early in metazoan evolution. Each METTL lineage originated from a single independent ancestor, forming monophyletic clades, indicating strong selective constraints driving their specialization. However, nucleoside-modifying and protein-modifying METTLs did not form unique monophyletic clades, and showed differences in their rates of evolution.

Summary: This study presents a ranking of all survival-related genes in chemotherapy treated basal and estrogen positive/HER2 negative breast cancer, providing a reference for future biomarker research. By analyzing hazard rates and p values of genes, the most promising prognostic genes can be identified, helping to focus research on candidates with the most clinical significance.
