Immunotherapy reversed myopathy but not cardiomyopathy in a necrotizing autoimmune myopathy patient with positive anti-SRP and MDA-5 autoantibodies

Xue Ma et al.

Summary: This study reported a NAM patient with both anti-SRP and MDA-5 antibodies, who showed significant improvement in skeletal muscle weakness after immunotherapy, but developed severe dilated cardiomyopathy 3 years later and eventually underwent heart transplantation. The patient remained symptom-free on oral prednisone and tacrolimus post-transplantation, suggesting a role of autophagy dysfunction and increased apoptosis in cardiac damage in anti-SRP and MDA5-positive NAM.

PERK signaling through C/EBPδ contributes to ER stress-induced expression of immunomodulatory and tumor promoting chemokines by cancer cells

Namratha Sheshadri et al.

Summary: ER stress in cancer cells can promote cell survival and aggressiveness, but prolonged stress can lead to cell death, offering a potential avenue for cancer therapies. The PERK pathway plays a role in regulating protein synthesis and promoting the secretion of tumor-promoting chemokines through transcription factor C/EBP delta. Further studies are needed to fully understand the impact of ER stress on tumor biology and its potential therapeutic implications.

Endoplasmic Reticulum-Plasma Membrane Contact Sites: Regulators, Mechanisms, and Physiological Functions

Chenlu Li et al.

Summary: Endoplasmic reticulum (ER) forms direct membrane contact sites with the plasma membrane (PM) in eukaryotic cells, playing crucial roles in lipid homeostasis, ion dynamics, and cell signaling through protein-protein or protein-lipid interactions. Distinct tethering factors dynamically regulate the architecture of ER-PM junctions in response to signals, and the physiological roles of these contact sites rely on various regulators acting in diverse cellular processes. This review focuses on proteins at ER-PM contact sites, highlighting recent progress in their mechanisms and physiological roles.

Crosstalk Between ER Stress, Autophagy and Inflammation

Sandhya Chipurupalli et al.

Summary: The endoplasmic reticulum (ER) is crucial for protein synthesis, folding, sensing cellular stress and maintaining cellular homeostasis. The unfolded protein response (UPR) expands protein folding capacity, reduces protein synthesis, enhances ER-associated protein degradation (ERAD), and amplifies cytokine-mediated inflammatory responses. UPR also activates autophagy, which limits inflammation and provides cyto-protection by attenuating ER-stress.

The Prognostic Value of PERK in Cancer and Its Relationship With Immune Cell Infiltration

Peng Wang et al.

Summary: The study found that PERK is differentially expressed in various cancers, with high expression associated with poor prognosis in KIRP, LGG, BRCA, and THCA, but favorable prognosis in HNSC. GSEA results indicated that PERK is mainly enriched in immune-related signaling pathways in BRCA, HNSC, and THCA. Additionally, PERK expression is significantly correlated with infiltrating levels of macrophages and dendritic cells, and with various immune markers, especially macrophage mannose receptor 1 (MRC1, also called CD206) and T-helper cells (Th).

Integrated signaling system under endoplasmic reticulum stress in eukaryotic microorganisms

Ting Cao et al.

Summary: The endoplasmic reticulum (ER) is a crucial multifunctional organelle responsible for correct folding and assembly of secretory and transmembrane proteins. Perturbations in ER function can lead to ER stress, activating the unfolded protein response (UPR) to cope with misfolded protein accumulation. If the imbalance cannot be restored, it may lead to cell death or diseases. The UPR is a coordination system that regulates transcription and translation to help cells recover ER homeostasis. The integrated signaling system in cells includes ER-associated degradation (ERAD), autophagy, hypoxia signaling, and mitochondrial biogenesis to cope with ER stress.
