Cancer Statistics, 2021

Rebecca L. Siegel et al.

Summary: Every year, the American Cancer Society projects the numbers of new cancer cases and deaths in the United States, with the latest data showing a significant decline in lung cancer mortality, while prostate cancer mortality has plateaued and breast and colorectal cancer mortality have slowed. Improvements in treatment have accelerated progress against lung cancer, leading to a record drop in overall cancer mortality.

Determinants of anti-PD-1 response and resistance in clear cell renal cell carcinoma

Lewis Au et al.

Summary: The ADAPTeR study aimed to investigate the mechanism of therapeutic response to nivolumab in metastatic clear cell renal cell carcinoma, with genomic analysis showing no correlation between tumor molecular features and response. Pre-existing immunity may play a key role in predicting clinical response.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Intratumoral CXCL13+CD8+T cell infiltration determines poor clinical outcomes and immunoevasive contexture in patients with clear cell renal cell carcinoma

Siyuan Dai et al.

Summary: This study revealed the unfavorable clinical outcomes of CXCL13(+)CD8(+)T cells in patients with ccRCC and their impairment of the immune function of total CD8(+)T cells. In addition, CXCL13(+)CD8(+)T cells also showed increased exhausted markers, decreased effector molecules, and better proliferation ability, associated with an immunoevasive tumor microenvironment.

Radiogenomics in Colorectal Cancer

Bogdan Badic et al.

Summary: Colorectal carcinoma is characterized by intratumoral heterogeneity that can be assessed by radiogenomics, which combines high-throughput quantitative data from medical imaging with molecular analysis. This approach is expected to advance personalized medicine but is still in its early stages with many challenges to overcome. The integration of image-derived features-radiomics and genomic profiles-genomics is a rapidly evolving field known as radiogenomics, with various studies dedicated to colorectal cancer showing potential for enhancing clinical decision-making.

Single-cell sequencing links multiregional immune landscapes and tissue-resident T cells in ccRCC to tumor topology and therapy efficacy

Chirag Krishna et al.

Summary: Clear cell renal cell carcinomas (ccRCCs) are highly immune infiltrated, and this study investigates the immune landscape of ccRCC using paired single-cell RNA (scRNA) and T cell receptor (TCR) sequencing, revealing extensive heterogeneity within and between patients. The study shows that tissue-resident T cells and tumor-associated macrophages (TAMs) are associated with responses to immune checkpoint blockade (ICB) and targeted therapies, highlighting the importance of cellular programs in therapeutic efficacy for ccRCC.

Transcriptional Profiling Reveals Kidney Neutrophil Heterogeneity in Both Healthy People and ccRCC Patients

Yiliang Meng et al.

Summary: This study utilized single-cell RNA sequencing to explore the subpopulation distribution of neutrophils in a healthy kidney and ccRCC, revealing different functional characteristics in these two environments. Neutrophils in ccRCC may exhibit resistance to immune therapies, suggesting potential challenges in using these treatments for this type of renal cancer.

Ferroptosis-Related Gene Signature Accurately Predicts Survival Outcomes in Patients With Clear-Cell Renal Cell Carcinoma

Kaili Chang et al.

Summary: A study on the expression data of ferroptosis-related genes in clear-cell renal cell carcinoma (ccRCC) revealed that a prognostic signature constructed by 11 ferroptosis genes was significantly related to overall survival and could predict prognosis accurately. The signature was found to be an independent prognostic factor and provide valuable insights for individualized treatment of ccRCC patients.
