Identification and Quantification of Necroptosis Landscape on Therapy and Prognosis in Kidney Renal Clear Cell Carcinoma

Sheng Xin et al.

Summary: In this study, the necroptosis landscape of KIRC patients was quantified and two distinct necroptosis-related patterns were identified. A stable and accurate prognostic marker called NRGscore was constructed, which showed significant correlations with clinicopathological characteristics, tumor immune microenvironment, and tumor mutation burden of KIRC patients. NRGscore also had effective guiding significance for immunotherapy, chemotherapy, and targeted drugs.

Copper induces cell death by targeting lipoylated TCA cycle proteins

Peter Tsvetkov et al.

Summary: Copper is an essential cofactor for all organisms, but excessive copper can lead to cell death. We found that copper-dependent cell death mechanism is different from known mechanisms, and it is related to mitochondrial respiration. Copper directly binds to lipoylated components of the tricarboxylic acid cycle, causing protein aggregation and loss of iron-sulfur cluster proteins, ultimately leading to cell death.

Roles of the Dynamic Tumor Immune Microenvironment in the Individualized Treatment of Advanced Clear Cell Renal Cell Carcinoma

Enyu Lin et al.

Summary: Immune checkpoint inhibitors are a first-line treatment option for ccRCC, but many patients do not respond to them, even with combination therapy. Studies have shown that the tumor immune microenvironment (TIME) plays a crucial role in the response to immunotherapy and targeted therapy in ccRCC patients, which can be influenced by targeted therapy and tumor genomic characteristics.

Comprehensive Analysis of the Immune Infiltrates of Pyroptosis in Kidney Renal Clear Cell Carcinoma

Zhuolun Sun et al.

Summary: This study systematically analyzed the role of pyroptosis in KIRC patients, establishing a predictive model that effectively differentiated between high- and low-risk patients and was validated in external cohorts. High-risk patients showed increased immune cell infiltration and higher ICI expression in the TME, leading to a poorer prognosis.

A Contemporary Review of Immune Checkpoint Inhibitors in Advanced Clear Cell Renal Cell Carcinoma

Eun-mi Yu et al.

Summary: The use of checkpoint inhibitors in advanced and metastatic renal cell carcinomas has advanced rapidly in recent years. Combination therapies with immune checkpoint inhibitors and VEGF inhibitors have further improved outcomes. Promising therapies are expected to make more breakthroughs in the future.
