Compositional, Structural, and Kinetic Aspects of Lipid Digestion and Bioavailability: In Vitro, In Vivo, and Modeling Approaches

Sebastien Marze

Summary: This article investigates the kinetics of lipid digestion and bioavailability and their impact on various diseases. The study finds a close link between lipid digestion kinetics and bioavailability kinetics, which can be modulated through food design or combination. The article presents different approaches, such as in vitro, modeling, in vivo, and mixed methods, to study the kinetics of both processes. It also proposes a unifying modeling scheme to better understand the role and interactions of various factors involved in lipid metabolism.

Oral delivery of therapeutic peptides and proteins: Technology landscape of lipid-based nanocarriers

Soheil Haddadzadegan et al.

Summary: The oral administration of therapeutic peptides and proteins faces various barriers, which can be overcome by using lipid-based nanocarriers. These nanocarriers enhance the lipophilic character of the drugs and protect them from enzymatic degradation. They can penetrate mucus and absorption membranes, enabling systemic delivery of the therapeutic molecules. Lipid-based nanocarriers show promising stability and delivery efficacy.

In vitro digestion of emulsified lard-based diacylglycerols

Xiaoqin Diao et al.

Summary: PGL showed higher hydrolysis rate and digestion extent than lard; zeta-potentials of the three lipids increased at first and then decreased during digestion; particle size of lard was consistently larger than that of GL and PGL throughout digestion.

Mathematical modelling of food hydrolysis during in vitro digestion: From single nutrient to complex foods in static and dynamic conditions

Steven Le Feunteun et al.

Summary: In vitro digestion methods and mathematical modeling are complementary tools for quantitatively comparing the impact of food composition and structure on the hydrolysis of macronutrients. Research focuses on modeling the hydrolysis of macronutrients at different digestive stages, and bridging the gap between in vitro and in vivo studies through modeling approaches is an ongoing effort. Hybrid strategies combining in vitro and in silico methods show promise in accurately translating in vitro observations into in vivo predictions.

Impact of gastrointestinal tract variability on oral drug absorption and pharmacokinetics: An UNGAP review

Zahari Vinarov et al.

Summary: Variability in the absorption of oral drugs can be attributed to differences in physiology, special populations, drug properties, and food-drug interactions. Pharmaceutical formulation approaches have been shown to reduce variability in oral drug exposure, but there are still gaps in understanding the impact of colonic factors, sex differences, and gut-related diseases on drug absorption variability.

Transient Permeation Enhancer® (TPE®) technology for oral delivery of octreotide: a technological evaluation

David J. Brayden et al.

Summary: The FDA approval of oral semaglutide for type 2 diabetes and oral octreotide for acromegaly demonstrates that selected niche peptides can be administered orally with the use of selected intestinal permeation enhancers. The oral octreotide formulation MYCAPSSA has shown promising results in terms of safety and efficacy, offering acromegaly patients a convenient alternative to monthly depot injections. The Transient Permeation Enhancer technology used in this formulation has shown potential in improving oral absorption of macromolecules.

INFOGEST inter-laboratory recommendations for assaying gastric and pancreatic lipases activities prior to in vitro digestion studies

Myriam M. L. Grundy et al.

Summary: In vitro digestion studies often use animal digestive enzyme extracts as substitutes of human gastric and pancreatic secretions, and measure the activities of enzymes using the pH-stat technique. While repeatability within individual laboratories is satisfactory, inter-laboratory variability is high.
