The horizon of bone organoid: A perspective on construction and application

Shuangshuang Chen et al.

Summary: The repair and regeneration of bone defects caused by various factors have always been a key issue in clinical practice. Bone tissue engineering technology has greatly accelerated the speed and quality of bone regeneration by providing functional scaffold materials and seed cells. However, the complicated bone metabolism mechanism poses challenges to research in bone regeneration. Organoids, as a new concept, show promising application prospects in organ development, drug screening, and mechanism study.

H19 Promotes HCC Bone Metastasis Through Reducing Osteoprotegerin Expression in a Protein Phosphatase 1 Catalytic Subunit Alpha/p38 Mitogen-Activated Protein Kinase-Dependent Manner and Sponging microRNA 200b-3p

Zhao Huang et al.

Summary: The study revealed that H19 plays a critical role in HCC bone metastasis by reducing OPG expression through the PPP1CA-induced inactivation of the p38MAPK pathway, and also functions as a sponge for miR-200b-3p to enhance cell migration and invasion.

RNF219/α-Catenin/LGALS3 Axis Promotes Hepatocellular Carcinoma Bone Metastasis and Associated Skeletal Complications

Shuxia Zhang et al.

Summary: The study reveals that HCC-secreted LGALS3 plays a crucial role in bone metastasis of HCC patients by promoting the formation of pre-metastatic niche and inducing skeletal-related events. Treatment with vertiporfin, a drug for macular degeneration, effectively inhibits skeletal complications of HCC.

Small extracellular vesicles deliver osteolytic effectors and mediate cancer-induced osteolysis in bone metastatic niche

Qinyu Ma et al.

Summary: The study reveals that EVs derived from prostate cancer cells deliver osteolytic signals to osteoclasts by carrying miR-152-3p, facilitating the osteolytic progression in bone metastasis.

Small Extracellular Vesicles and Metastasis-Blame the Messenger

Tanja Seibold et al.

Summary: Metastatic lesions in cancer pose challenges for treatment, as tumor cells communicate with local and distant cells via sEVs to promote metastasis. sEVs reprogram target cells by delivering bioactive cargos, influencing tumor growth, cell motility, angiogenesis, and immune evasion.

Trabecular bone organoids: a micron-scale 'humanised' prototype designed to study the effects of microgravity and degeneration

Alexandra Iordachescu et al.

Summary: The authors developed a micron-scale bone organoid prototype to study bone processes in microgravity, observing phenomena and functions of bone marrow cells and bone cells. The research results indicate that the locations of osteoclastic bone resorption differ in morphology between the simulated microgravity group and static group.

Pre-metastatic Niche Formation in Different Organs Induced by Tumor Extracellular Vesicles

Qi Dong et al.

Summary: Primary tumors modify the microenvironment of distant organs to create a supportive pre-metastatic niche, facilitating metastasis. Extracellular vesicles play a critical role in PMN formation by delivering functional molecules to recipient cells in target organs.
