Tumor IsomiR Encyclopedia (TIE): a pan-cancer database of miRNA isoforms

Xavier Bofill-De Ros et al.

Summary: MicroRNAs (miRNAs) play a key role in regulating gene expression in cancers, with their sequence variants or isoforms (isomiRs) possessing unique functions. The Tumor IsomiR Encyclopedia (TIE) is a dynamic database that aggregates isomiR variants from over 10,000 tumor samples in TCGA and TARGET projects, addressing challenges in mapping isomiRs and overcoming the disregard of low frequency events. TIE's ability to annotate heterogeneous isomiR sequences and provide results for online browsing or download showcases its utility in analyzing miR-21 and miR-30a isomiRs.

miRGTF-net: Integrative miRNA-gene-TF network analysis reveals key drivers of breast cancer recurrence

Stepan Nersisyan et al.

Summary: Analysis of regulatory networks using miRGTF-net is effective in identifying and quantifying intracellular interactions. The tool can construct reliable prognostic gene signatures for ER-positive breast cancer using paired miRNA/mRNA-sequencing data. An essential part of these prognostic genes are direct targets of transcription factor E2F1, indicating a potential interaction between estrogen receptor alpha and E2F1 in cancer recurrence.

Hairpin sequence and structure is associated with features of isomiR biogenesis

Anton Zhiyanov et al.

Summary: MiRNA isoforms (isomiRs) are single-stranded small RNAs produced from the same pri-miRNA hairpin through cleavage by Drosha and Dicer enzymes. Analysis of miRNA processing positions across 31 different cancers revealed that the positions are not tissue-specific and a model based on surrounding nucleotides can distinguish between homogeneous and heterogeneous cleavage. Additionally, some miRNAs show imprecise Dicer cleavage at the 5' end, while almost all cleavage sites at the 3' end are heterogeneous.

Hypoxia-Induced miR-148a Downregulation Contributes to Poor Survival in Colorectal Cancer

Stepan Nersisyan et al.

Summary: Hypoxia is extensively studied for its impact on various pathophysiological processes such as cancer progression. The contribution of miRNAs to molecular response to hypoxia varies significantly depending on cell types and causes of hypoxia. The regulatory network induced by hypoxia is complex and plays a crucial role in tumor progression.
