Combined Network Pharmacology and Cytology Experiments to Identify Potential Anti-Breast Cancer Targets and Mechanisms of Delphinidin

Jiayuan Peng et al.

Summary: Delphinidin, a type of anthocyanin monomer, has shown potential anti-breast cancer effects through comprehensive network pharmacology, cellular and molecular experiments. Molecular docking validated important core targets (such as EGFR, TOP2A, and PTGS2) and suggested delphinidin may offer effective approaches in breast cancer prevention and therapy.

Statistics and network-based approaches to identify molecular mechanisms that drive the progression of breast cancer

Shahin Alam et al.

Summary: This study identified differentially expressed genes (DEGs) between breast cancer (BC) and control samples, constructed a protein-protein interaction network, and selected key genes. The DEGs were found to significantly enrich key biological processes, molecular functions, cellular components, and pathways related to BC, and had strong prognostic power. Transcriptional and post-transcriptional regulators of these key genes were also detected. Additionally, three repurposable candidate-drugs for BC treatment were suggested and validated through molecular docking analysis.

Inhibition of the PINK1-Parkin Pathway Enhances the Lethality of Sorafenib and Regorafenib in Hepatocellular Carcinoma

Shun Zhang et al.

Summary: This study found that sorafenib and regorafenib induce cell death in hepatocellular carcinoma by damaging mitochondria, and mitophagy can alleviate this cell death. Inhibiting autophagy or mitochondrial fission can aggravate the cell death caused by these drugs. Additionally, knocking down the protein PINK1 can enhance the inhibitory effects of sorafenib and regorafenib on hepatocellular carcinoma.

Novel Dual PI3K/mTOR Inhibitor, Apitolisib (GDC-0980), Inhibits Growth and Induces Apoptosis in Human Glioblastoma Cells

Wioleta Justyna Omeljaniuk et al.

Summary: The study found that GDC-0980 can effectively inhibit the survival of human GBM cells and induce apoptosis, independent of ER stress-mediated DR5 activation. It is suggested that GDC-0980 may block the inhibitory effect on protein synthesis through inhibiting PERK expression, leading to an intensification of translation and an increase in apoptosis. Additionally, CHOP stimulates protein synthesis and increases apoptosis.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Xiao Xiao et al.

Summary: This study identified potential pivotal genes for prognosis and treatment in TNBC, with CCNB1, NCAPG, MCM4, and RRM2 found to be associated with poor prognosis in TNBC and may act as promising prognostic biomarkers and therapeutic targets. Functional analysis revealed the involvement of these genes in tumor proliferation.

Masitinib is a broad coronavirus 3CL inhibitor that blocks replication of SARS-CoV-2

Nir Drayman et al.

Summary: This study identified a drug called masitinib that can inhibit the replication of both SARS-CoV-2 and OC43 virus, and acts by competitively inhibiting 3CLpro. The drug showed strong antiviral effects in in vivo experiments on mice infected with SARS-CoV-2, and was effective against multiple variants in vitro.
