Calpain-2 regulates hypoxia/HIF-induced plasticity toward amoeboid cancer cell migration and metastasis

Veronika te Boekhorst et al.

Summary: This article studies the molecular mechanisms of cancer cell invasion and metastasis under hypoxic conditions and identifies calpain-2 as a key regulator of the transition from collective to amoeboid dissemination in breast and head and neck carcinoma cells. The study reveals that hypoxia induces calpain-2-mediated amoeboid conversion by deactivating beta 1 integrins. Targeted intervention of calpain-2 activity can restore normal cell phenotype and prevent lung metastasis.

Nucleation, stabilization, and disassembly of branched actin networks

Alexis M. Gautreau et al.

Summary: This article discusses the role and regulation of the Arp2/3 complex, a nucleation and branching machinery of actin filaments conserved in all eukaryotes. Recent advances include high-resolution cryo-EM structures that provide insights into the mechanisms of complex activation and branch formation, as well as novel cellular pathways for branch formation, stabilization, and debranching. However, there are still significant gaps in our understanding of complex inhibition and branch stabilization and disassembly.

Emerin regulation of nuclear stiffness is required for fast amoeboid migration in confined environments

Sandrine B. Lavenus et al.

Summary: It has been found that increasing nuclear stiffness through elevating lamin A levels inhibits fast amoeboid migration in melanoma cells. The inner nuclear membrane protein emerin plays a key role in regulating this migration process, and its activity needs to be precisely calibrated. Interestingly, low activity of Src has been observed in cells within a confined environment, which is related to fast amoeboid migration.

Vimentin intermediate filaments and filamentous actin form unexpected interpenetrating networks that redefine the cell cortex

Huayin Wu et al.

Summary: The cytoskeleton of eukaryotic cells consists of various filamentous proteins, including F-actin, microtubules, and intermediate filaments. Interactions between these cytoskeletal components play a crucial role in determining cell structure and function. This study investigates the interplay between F-actin and vimentin intermediate filament (VIF) networks, revealing their synergistic effects in both structure and function. These findings significantly broaden our understanding of the contributions of the cytoskeleton components, particularly the interactions between intermediate filaments and F-actin.

Summary: Vimentin, in addition to contributing to the mechanical stabilization of cell structure, also helps to control the assembly of cell adhesions and migration through collagen matrices. It plays a central regulatory role in the assembly of focal adhesions and is a key organizer of the beta 1 integrin adhesive machinery, affecting cell migration through collagen.
