T-Cell-Specific CerS4 Depletion Prolonged Inflammation and Enhanced Tumor Burden in the AOM/DSS-Induced CAC Model

Summary: This study reveals the importance of CerS4 in the multifactorial process of inflammatory bowel disease (IBD) and colitis-associated cancer (CAC). Deficiency of CerS4 in T-cells leads to sustained activation and prevents the resolution of inflammation, resulting in a higher tumor burden in a mouse model of CAC. In contrast, CerS4 deficiency in epithelial cells leads to smaller colon tumors and appears to be beneficial.

Glucosylceramide in T cells regulates the pathology of inflammatory bowel disease

Summary: Inflammatory bowel disease (IBD) is a chronic inflammatory disease characterized by excessive activation of T cells. This study investigated the involvement of glycosphingolipids (GSLs) in IBD. The results showed that decreased expression of UDPglucose ceramide glucosyltransferase (UGCG), a GSLs synthase, was associated with IBD. Decreased UGCG led to worsened colitis and lower levels of regulatory T cells (Treg). Additionally, treatment with glucosylceramide (GlcCer) nanoparticles showed potential efficacy in preventing DSS-induced colitis, suggesting GlcCer in T cells as a therapeutic target for IBD patients.

Ganglioside GM3 Synthase Deficiency in Mouse Models and Human Patients

Summary: Gangliosides are a type of glycosphingolipids highly expressed in neural tissues, and GM3 synthase is an enzyme responsible for their synthesis. Mutations in the ST3GAL5 gene, which encodes GM3 synthase, cause severe neurological diseases in humans. Knockout mice lacking GM3 synthase exhibit hearing impairment and enhanced insulin sensitivity, but do not display the neurological defects seen in human patients.

Summary: The pathobiology of rheumatoid inflammatory diseases involving innate and adaptive immune components and synoviocytes is still not fully understood. Single-cell analyses of patient samples and relevant mouse models have characterized many cellular subsets in rheumatoid arthritis (RA). This study revealed that murine macrophage subtypes, similar to those found in RA patients, were expanded in arthritis and linked to promoting the function of Th17 cells in the joint. In vitro experiments further identified a cellular feedback loop involving TNF-alpha that promotes Th17 cell pathogenicity and drives inflammatory arthritis.
