Summary: This study found that exogenous supplementation of itaconate inhibits inflammation and senescence of chondrocytes through the Nrf2/STING/NF-kappa B axis, reducing the progression of osteoarthritis. Itaconate also regulates the polarization of macrophages, reducing apoptosis of chondrocytes. In vivo, intra-articular injection of itaconate reduces cartilage degradation and synovial membrane inflammation in a mouse model of osteoarthritis.

Summary: This study demonstrated that exercise-induced Irisin has anti-inflammatory and antipyroptosis effects on osteoarthritis chondrocytes. Moderate-intensity treadmill exercise increased the levels of Irisin, which recovered the expression of collagen II and attenuated the expression of inflammatory markers. Irisin achieved these effects by inhibiting the PI3K/Akt/NF-kappa B signaling pathway and inhibiting the activity of NLRP3/caspase-1. The findings suggest that Irisin could be a potential therapeutic target for osteoarthritis.
