Da-Chai-Hu-Tang Protects From Acute Intrahepatic Cholestasis by Inhibiting Hepatic Inflammation and Bile Accumulation via Activation of PPARα

Summary: The study demonstrates that Da-Chai-Hu-Tang (DCHT) can prevent acute intrahepatic cholestasis and liver injury, but is ineffective against extrahepatic cholestasis. DCHT restores bile acid homeostasis and improves glutathione imbalance, thereby alleviating liver inflammation caused by intrahepatic cholestasis. Compounds in DCHT with high affinity for PPAR alpha mimic its cytoprotective effect on intrahepatic cholestasis.

Summary: The study found that the plasma ratio of UDC-Tau to 7oxoLC-Tau can serve as a biomarker for decreased 11 beta-HSD1 oxoreduction activity in different mouse models. Enzyme product to substrate ratios were more reliable markers of 11 beta-HSD1 activity than absolute levels due to large inter-individual variations in bile acid concentrations. The persistence of oxoreduction activity in the face of cofactor loss indicates the existence of an alternative NADPH source in the endoplasmic reticulum.

Hyocholic acid species improve glucose homeostasis through a distinct TGR5 and FXR signaling mechanism

Summary: This study reveals that high levels of hyocholic acid (HCA) in pigs contribute to their resistance against type 2 diabetes, as depletion of bile acids (BA) in pigs leads to reduced glucagon-like peptide-1 (GLP-1) secretion and increased blood glucose levels. HCA administration in diabetic mouse models effectively improves GLP-1 secretion and glucose homeostasis by activating TGR5 and inhibiting FXR in enteroendocrine cells, a mechanism not seen in other BA species. Clinical findings also show a correlation between lower levels of HCA species and diabetes, indicating its importance in glycemic control.

Inflammation-induced cholestasis in cancer cachexia

Natural and synthetic FXR agonists, as well as FXR-regulated factors like fibroblast growth factor 19 (FGF19), are being evaluated as potential treatments for metabolic diseases such as type 2 diabetes and non-alcoholic fatty liver disease.
