Effect of the Size of Protein Therapeutics on Brain Pharmacokinetics Following Systematic Administration

Summary: In this study, the impact of protein therapeutic size on brain pharmacokinetics (PK) following systemic administration in rats was investigated. The results showed that the exposure of protein therapeutics in different brain regions could be significantly different, and there could be optimal sizes of protein therapeutics to achieve maximum/selective exposure.

A Novel EGFRvIII T-Cell Bispecific Antibody for the Treatment of Glioblastoma

Summary: T-cell bispecific antibodies (TCB) are engineered molecules that can bind both the T-cell receptor and tumor-specific antigens, showing potential therapeutic effects for EGFRvIII mutation in glioblastoma (GBM). We designed and developed a novel EGFRvIII-TCB with specificity and potent antitumor activity, promoting T-cell activation and cytokine secretion for tumor cell killing, as well as T-cell recruitment into tumors. In GBM animal models, including humanized orthotopic patient-derived xenograft models, EGFRvIII-TCB induced tumor regression. These results support the clinical testing of EGFRvIII-TCB.

Pharmacokinetics and Pharmacodynamics of T-Cell Bispecifics in the Tumour Interstitial Fluid

Summary: This study investigated the pharmacokinetics and cytokine release of two T-cell bispecific antibodies with different binding affinities in a tumour-bearing humanised mouse model. High tumour interstitial concentrations were observed for both antibodies, influenced by their respective target binding affinities. This tailored experimental method and mathematical model can provide insights into the impact of target affinity on tumour accumulation and PKPD assessment of therapeutic antibodies.

Collecting antibodies and large molecule biomarkers in mouse interstitial brain fluid: a comparison of microdialysis and cerebral open flow microperfusion

Summary: This study compared two methods, microdialysis and cerebral open flow microperfusion, for measuring the concentration of large molecules in the interstitial brain fluid in mice. Without recovery correction, concentrations from cerebral open flow microperfusion were over 10 times higher than those from microdialysis. The use of in vivo recovery revealed a 6-fold higher absolute concentration at steady state compared to calculations based on in vitro recovery. Technical optimization has improved the performance of both systems, allowing for the simultaneous sampling of up to 12 mice.

Brain pharmacokinetics of anti-transferrin receptor antibody affinity variants in rats determined using microdialysis

Summary: Receptor-mediated transcytosis (RMT) is used to enhance the delivery of monoclonal antibodies (mAb) into the central nervous system (CNS). The binding affinity of mAbs has a complex relationship with pharmacokinetics (PK) in the brain, with optimal dissociation constant (K-D) values resulting in the highest brain exposure of mAbs in the brain interstitial fluid (bISF). This study highlights the importance of accurately measuring the PK of CNS-targeted antibodies using microdialysis in the brain.
