Decyl esters production from soybean-based oils catalyzed by lipase immobilized on differently functionalized rice husk silica and their characterization as potential biolubricants

Summary: This study aimed to produce decyl esters through a two-step process of hydrolysis and esterification. High hydrolysis rates were achieved by using Candida rugosa enzyme for the hydrolysis of refined soybean oil and used soybean oils. Efficient esterification of decyl esters in solvent-free systems was achieved using Thermomyces lanuginosus immobilized on silica particles extracted from rice husk. Octyl-SiO2 was found to be the most stable immobilization support after fifteen successive esterification batches.

An Engineered Cytidine Deaminase for Biocatalytic Production of a Key Intermediate of the Covid-19 Antiviral Molnupiravir

Summary: The Covid-19 pandemic has emphasized the need for efficient and cost-effective production of antiviral drugs on a large scale. This study presents a concise biocatalytic process for synthesizing Molnupiravir, a nucleoside analogue used for oral treatment of SARS-CoV-2. The novel biocatalyst developed through evolutionary adaptation of the enzyme cytidine deaminase exhibits remarkable efficiency, achieving over 85,000 turnovers within 3 hours and operating at high substrate loading. Additionally, in situ crystallization of the N-hydroxy-cytidine product increases its yield and facilitates its conversion to Molnupiravir.

Biocatalytic decarboxylative Michael addition for synthesis of 1,4-benzoxazinone derivatives

Summary: This study reports the Candida antarctica lipase B (Novozym 435) catalyzed novel decarboxylative Michael addition in vinylogous carbamate systems for the synthesis of 1,4-benzoxazinone derivatives. The reaction proceeds through Michael addition, ester hydrolysis, and decarboxylation. A possible mechanism involving simultaneous lipase-catalyzed Michael addition and ester hydrolysis is suggested. The reaction demonstrates enzymatic promiscuity by offering formation of complex products through multi-step reactions in a one pot process under mild and facile reaction conditions with moderate to high yields and no side product formation.

Summary: Trimethylolpropane triesters (TMPTEs) were produced through enzymatic hydroesterification of refined soybean oil (RSO) or used soybean cooking oil (USCO). Immobilized Eversa (R) Transform 2.0 showed the highest activity, retaining above 90% of its original activity after 13 consecutive reaction cycles. The produced TMPTEs exhibited good cold temperature properties with pour point values between -11 and -9 degrees C.

Summary: In this study, melatonin and ferulic acid were incorporated into tertiary amide-based histone deacetylase 6 (HDAC6) inhibitors to develop multi-target-directed inhibitors for neurodegenerative diseases, maintaining antioxidant effects while ensuring high affinity and selectivity at HDAC6. The resulting compound 10b showed significant and selective inhibition of HDAC6, comparable antioxidant abilities to ferulic acid and melatonin, and no neurotoxicity. It also exhibited immunomodulatory effects and demonstrated higher efficacy in an AD mouse model compared to the positive control HDAC6 inhibitor ACY1215 and an equimolar mixture of ACY1215, melatonin, and ferulic acid, suggesting potential disease-modifying properties.
