Summary: Articular cartilage repair remains a challenging task and tissue engineering has been widely employed in joint tissue reconstruction. However, the optimal scaffolds for cartilage repair are still unknown. This study compared five different scaffolds and found that decellularized cartilage and collagen type II-based scaffold almost fully restored the congruence of the articular surface. However, the formation of hyaline cartilage was not observed in any group. It was concluded that for restoration of hyaline cartilage, scaffolds need to be combined with cellular therapy or morphogens promoting chondrogenic differentiation.

An additive manufacturing-based 3D printed poly e-caprolactone/alginate sulfate/extracellular matrix construct for nasal cartilage regeneration

Summary: This study used fused deposition modeling method to fabricate a poly e-caprolactone (PCL) scaffold, and filled it with Alginate (Alg): Alginate-Sulfate (Alg-Sul) hydrogel to create a biomimetic environment and mimic the structure of glycosaminoglycans. Different concentrations of decellularized extracellular matrix (dECM) were added to enhance chondrogenesis. Cellular analyses showed that the scaffold with 1% dECM exhibited better viscosity, cell viability, and proliferation. Mechanical characterization and chondrogenic differentiation tests demonstrated that the optimized scaffold (1% dECM) had similar mechanical properties to native nasal cartilage tissue and appropriate biochemical features.

Bioprinting of Chondrocyte Stem Cell Co-Cultures for Auricular Cartilage Regeneration

Summary: Advancements in 3D bioprinting technology have allowed for the controlled deposition of cells, leading to the development of constructs that match the anatomical features of patients. In this study, a 3D bioprinted scaffold made from GelMA/HAMA was used to investigate the monocultures and co-cultures of human septal chondrocytes and human bone marrow-derived mesenchymal stem cells. The findings demonstrated that the combination of these cell types produced cell proliferation and chondrogenic expressions that mimicked the characteristics of both cell types. This research suggests that co-cultures of BM-hMSCs and healthy septal PCs can be utilized to replace monocultures in cartilage regeneration studies.

The clinical potential of articular cartilage-derived progenitor cells: a systematic review

These progenitor cells, called articular cartilage-derived progenitor cells (ACPCs), can be isolated from cartilage of various species including humans, horses, and cows. ACPCs possess mesenchymal stromal cell (MSC)-like characteristics and show no signs of hypertrophic differentiation, making them promising for cartilage repair. However, there is currently no established unique cell marker for identifying ACPCs, leading to variations in isolation and characterization protocols.
