PatDREB Transcription Factor Activates Patchoulol Synthase Gene Promoter and Positively Regulates Jasmonate-Induced Patchoulol Biosynthesis

Summary: In the synthesis of patchoulol, the promoter of the patchoulol synthase gene (PatPTSpro) is induced by methyl jasmonate (MeJA). The nucleus-localized AP2/ERF transcription factor PatDREB acts as a transcription activator and binds to PatPTSpro, regulating patchoulol biosynthesis by modulating gene expression. PatDREB also interacts with jasmonate ZIM-domain 4 (JAZ4) and the MYB-related transcription factor PatSWC4, synergistically facilitating patchoulol biosynthesis. However, the transcriptional activation activity of the PatDREB-PatSWC4 complex can be inhibited by PatJAZ4, which can be reversed by JA.

Chromosome-level and haplotype-resolved genome provides insight into the tetraploid hybrid origin of patchouli

Summary: Translation: The ploidy level and evolutionary history of patchouli, an important aromatic plant, have been elucidated through genome assembly and analysis. Patchouli is found to be a tetraploid hybrid with compensated aneuploidy. Tandem duplication of sesquiterpene biosynthetic genes is a major contributor to the biosynthesis of patchouli bioactivity components. The genome is phased into two subgenomes, and a chromosome substitution event is identified. The divergence between subgenomes is caused by a burst of universal LTR-RTs in one subgenome. The evolutionary scenario of patchouli, including whole genome tetraploidization, subgenome divergency, hybridization, and chromosome substitution, is revealed.

The transcription factor OpWRKY2 positively regulates the biosynthesis of the anticancer drug camptothecin in Ophiorrhiza pumila

Summary: The limited bioavailability of plant-derived natural products with anticancer activity poses challenges to the pharmaceutical industry. Recent study reveals that OpWRKY2 acts as a direct positive regulator of camptothecin biosynthesis, which can increase camptothecin levels by more than three-fold. This finding provides a feasible strategy for the over-accumulation of camptothecin in a biotechnologically amenable system.

Transcriptome-Wide Identification of WRKY Transcription Factor and Functional Characterization of RgWRKY37 Involved in Acteoside Biosynthesis in Rehmannia glutinosa

Summary: In this study, 37 putative WRKY transcription factors were identified in Rehmannia glutinosa, and their structural features and transcriptional profiles under different treatments were analyzed. It was suggested that several RgWRKY members are involved in acteoside biosynthesis, with one specific gene, RgWRKY37, being selected for further functional research. These results strongly indicate the regulatory role of WRKY transcription factors in acteoside biosynthesis.

AaWRKY17, a positive regulator of artemisinin biosynthesis, is involved in resistance to Pseudomonas syringae in Artemisia annua

Summary: In this study, a novel WRKY transcription factor AaWRKY17 was identified in Artemisia annua, which could increase artemisinin content and resistance to Pseudomonas syringae. AaWRKY17 directly bound to the promoter region of ADS gene and promoted its expression, leading to increased levels of defense marker genes. Overexpression of AaWRKY17 in A. annua resulted in enhanced pathogen tolerance and artemisinin content. This suggests that AaWRKY17 could be used in transgenic breeding to improve artemisinin production and pathogen tolerance in A. annua.

The WRKY transcription factor AaGSW2 promotes glandular trichome initiation in Artemisia annua

Overexpression of a specific transcription factor in A. annua significantly increased glandular secreting trichome density, demonstrating a potential strategy to enhance the production of valuable secondary metabolites like artemisinin.
