In vivo enzymatic digestion of HRV 3C protease cleavage sites-containing proteins produced in a silkworm-baculovirus expression system

Jian Xu et al.

Summary: The Baculovirus expression vector system (BEVS) is an effective protein expression system and has been used to engineer important enzymes and vaccines. The use of fusion tags for protein purification can negatively affect the biological activity of the target protein. This study validated the cleavage activity of the human rhinoviruses (HRV) 3C protease in both Escherichia coli and silkworm-BEVS systems. The findings demonstrate the successful establishment of a tag-free protein engineering platform using a coinfection-based recognition-and-reaction BEVS system.

The recombinant Erns and truncated E2-based indirect enzyme-linked immunosorbent assays to distinguishably test specific antibodies against classical swine fever virus and bovine viral diarrhea virus

Weicheng Yi et al.

Summary: In this study, highly specific and sensitive ELISAs were developed for the serological diagnosis and surveillance of classical swine fever virus (CSFV) and bovine viral diarrhea virus (BVDV) infections. The ELISAs showed no cross-reaction and high sensitivity and specificity when tested with virus specific sera from pigs, immunized rabbits, and immunized mice. These newly developed ELISAs could be valuable tools for the serological diagnosis of CSFV and BVDV infections.

Immunogenicity and protective potential of chimeric virus-like particles containing SARS-CoV-2 spike and H5N1 matrix 1 proteins

Jing Chen et al.

Summary: This study designed and prepared four kinds of virus-like particles (VLPs) using an insect expression system. The results showed that the vaccine based on mSM and Al/CpG adjuvant could stimulate animals to produce sustained high-level antibodies and establish an effective protective barrier, suggesting its potential in preventing the COVID-19 pandemic.

Co-administration of a plasmid encoding CD40 or CD63 enhances the immune responses to a DNA vaccine against bovine viral diarrhea virus in mice

Dongze Leng et al.

Summary: This study investigated the adjuvant effect of CD40 and CD63 plasmids on the immune responses to a BVDV E2 DNA vaccine in mice. The results showed that plasmids encoding CD40 or CD63 significantly enhanced the immune responses to BVDV, including increased neutralizing antibody titers, anti-E2 IgG2a antibody levels, and lymphocyte proliferation and interferon-γ production.
