Protein acetylation regulates xylose metabolism during adaptation of Saccharomyces cerevisiae

Yong-Shui Tan et al.

Summary: This study constructed a xylose-utilizing S. cerevisiae for adaptation study, finding that the xylose consumption rate increased significantly in the second incubation compared with the first incubation. It was observed at the single-cell level that the stagnation time for xylose utilization decreased after adaptation with xylose medium. The study also identified H4K5 acetylation and genes GCN5 and HPA2 as important factors related to xylose consumption memory in engineered S. cerevisiae during adaptation.

Crabtree/Warburg-like aerobic xylose fermentation by engineered Saccharomyces cerevisiae

Sae-Byuk Lee et al.

Summary: The efficient conversion of xylose into biofuels has been limited by bottlenecks, but a new method involving engineered yeast strains has shown promise in aerobically fermenting xylose to ethanol. Through gene duplications and deletions, the rates and yields of xylose fermentation were increased, potentially enabling sustainable production of industrial biofuels. This research offers insights into how key regulatory mutations can prime yeast for aerobic xylose fermentation, leading to increased xylose flux and redirection into fermentation products.

Transcriptomic Changes Induced by Deletion of Transcriptional Regulator GCR2 on Pentose Sugar Metabolism in Saccharomyces cerevisiae

Minhye Shin et al.

Summary: The deletion of GCR2 gene has been identified to have a significant impact on xylose fermentation in Saccharomyces cerevisiae, leading to the upregulation of PPP genes and downregulation of glycolytic genes, particularly under xylose conditions. Although there was no synergistic effect between Delta pho13 and Delta gcr2 in improving xylose fermentation, the results suggest that GCR2 is a novel knockout target for enhancing lignocellulosic ethanol production.

Exploring metal ion metabolisms to improve xylose fermentation in Saccharomyces cerevisiae

Gisele Cristina de Lima Palermo et al.

Summary: In this study, alternative metabolisms involved in metal homeostasis were explored to enhance xylose consumption in engineered C5-strains of Saccharomyces cerevisiae. The vacuolar Fe2+/Mn2+ transporter CCC1 and the protein involved in heavy metal ion homeostasis BSD2 were identified as promising targets for rational metabolic engineering strategies. Results showed significant improvements in xylose consumption rates compared to control, suggesting a potential for enhancing biomass-derived sugar conversion in lignocellulose-based biorefineries.

Metabolic network remodelling enhances yeast's fitness on xylose using aerobic glycolysis

Xiaowei Li et al.

Summary: In this study, a combination of metabolic engineering, systems biology, and adaptive laboratory evolution was used to gain insights into how yeast rewires cellular processes to efficiently adapt to metabolic transitions from glucose to xylose. Through metabolic engineering, enhanced cell growth on xylose was achieved, with transcriptome analysis revealing involvement of multiple pathways in cellular reprogramming. Genome resequencing and reverse engineering identified key factors like SWI/SNF chromatin remodeling, osmotic response, and aldehyde reductase in improving growth on xylose, with glycerol-3-phosphate and xylitol identified as key metabolites affecting cellular adaptation.
