Augmin-dependent microtubule self-organization drives kinetochore fiber maturation in mammals

Summary: Chromosome segregation in mammals relies on the maturation of K-fibers, and augmin plays a key role in this process. Our study reveals that augmin promotes turnover and poleward flux of kinetochore and interpolar microtubules and increases the frequency of microtubule growth through branched microtubule nucleation. These findings are important for understanding the self-organization and maturation of K-fibers in mammals.

Length-dependent poleward flux of sister kinetochore fibers promotes chromosome alignment

Summary: This study discovered the importance of chromosome alignment at the spindle equator for proper chromosome segregation, which relies on the pulling forces exerted at kinetochore fiber tips and polar ejection forces. Additionally, the study found that the forces driving the poleward flux of kinetochore fibers also affect chromosome alignment. A flux-driven centering model was introduced, which depends on the flux generated by forces within the overlaps of bridging and kinetochore fibers. The study confirmed the key prediction that kinetochore fiber flux is length dependent. Several regulators of overlap and flux were identified, including Kif18A, Kif4A, and NuMA.

Three-dimensional structure of kinetochore-fibers in human mitotic spindles

Summary: In this study, the three-dimensional structure of kinetochore fibers in mammalian cells was characterized using large-scale electron tomography. It was found that each kinetochore fiber is composed of approximately nine kinetochore microtubules, with variations in circumference and density along their length. The association between kinetochore microtubules and non-kinetochore microtubules was primarily observed in the spindle pole regions.
