Comparative genomics reveals electron transfer and syntrophic mechanisms differentiating methanotrophic and methanogenic archaea

Grayson L. Chadwick et al.

Summary: The anaerobic oxidation of methane coupled to sulfate reduction is a microbial process that involves a partnership between anaerobic methanotrophic archaea and sulfate-reducing bacteria. This study reconstructed and analyzed 28 ANME genomes, enabling a comparative analysis of all marine ANME clades. The findings suggest that methane production may have evolved from a methanotrophic ancestor.

A 300-fold conductivity increase in microbial cytochrome nanowires due to temperature-induced restructuring of hydrogen bonding networks

Peter J. Dahl et al.

Summary: Geobacter sulfurreducens transports respiratory electrons over micrometers through nanowires composed of polymerized cytochrome OmcS. Individual nanowires exhibit hopping conductance and show a significant increase in their intrinsic conductance upon cooling. The cooling effect is caused by the rearrangement of hydrogen bonding networks in the nanowires, resulting in a temperature-sensitive switch for charge transport controlled by the protein surrounding the hemes.

A New Paradigm of Multiheme Cytochrome Evolution by Grafting and Pruning Protein Modules

Ricardo Soares et al.

Summary: This study investigates the evolution of multiheme cytochromes and reveals that NrfA and cytochrome c(554) belong to different clades, suggesting that they originated from truncation of ancestral octaheme cytochromes related to extant octaheme nitrite reductase and MccA, respectively. The research provides new insights into the evolutionary pathways of multiheme cytochromes and their role in the interplay between biology and geochemistry throughout Earth's history.

Cryo-EM structure of an extracellular Geobacter OmcE cytochrome filament reveals tetrahaem packing

Fengbin Wang et al.

Summary: Two electrically conductive appendages are produced by the bacterium Geobacter sulfurreducens. One is a conductive fiber containing polymerized c-type cytochrome OmcS subunits, and the other is a thinner appendage comprised of cytochrome OmcE subunits. Both filaments share a conserved haem packing arrangement in which the haems are coordinated by histidines in adjacent subunits, despite having no overall sequence or structural similarities.

Proteolytic Maturation of the Outer Membrane c-Type Cytochrome OmcZ by a Subtilisin-Like Serine Protease Is Essential for Optimal Current Production by Geobacter sulfurreducens

Ayako Kai et al.

Summary: The c-type cytochrome OmcZ in Geobacter sulfurreducens is essential for optimal current production in microbial fuel cells. A subtilisin-like serine protease called OzpA plays a key role in cleaving OmcZ and generating the mature form OmcZ(S), which is crucial for extracellular electron transfer to electrodes. This proteolytic maturation of OmcZ is a unique regulation mechanism in G. sulfurreducens, providing new insights for efficient energy conversion from chemical energy to electricity.

Highly accurate protein structure prediction with AlphaFold

John Jumper et al.

Summary: Proteins are essential for life, and accurate prediction of their structures is a crucial research problem. Current experimental methods are time-consuming, highlighting the need for accurate computational approaches to address the gap in structural coverage. Despite recent progress, existing methods fall short of atomic accuracy in protein structure prediction.
