Respiratory Influenza Virus Infection Causes Dynamic Tuft Cell and Innate Lymphoid Cell Changes in the Small Intestine

Shanley N. Roach et al.

Summary: Influenza A viruses (IAV) can cause severe disease and death in humans. In this study, it was found that respiratory IAV infection leads to dynamic changes in tuft cells and ILCs in the small intestines. Tuft cells were found to be necessary for the infection-induced increase in small intestine ILC2s. These findings suggest that the intestinal changes in tuft cell and ILC populations may play a role in preventing systemic infection and contribute to severe disease in humans with preexisting conditions.

IL-13-programmed airway tuft cells produce PGE2, which promotes CFTR-dependent mucociliary function

Maya E. Kotas et al.

Summary: Chronic type 2 inflammatory diseases of the respiratory tract are characterized by mucus overproduction and disordered mucociliary function. Common epithelial cell types (mucus secretory and ciliated cells) are affected by IL-13, but it is unclear what role rare cells (tuft cells) play in airway T2 inflammation. This study found that IL-13 expands and programs airway tuft cells towards eicosanoid metabolism and deficiency of tuft cells leads to reduced airway PGE concentration. Tuft cells have a role in regulating epithelial mucociliary function in the allergic airway.

Summary: Single-nucleus RNA sequencing of lung tissues from individuals who died of COVID-19 revealed a highly inflamed lung environment with impaired T cell responses and failed transition of alveolar type 2 cells. Additionally, expansion of pathological fibroblasts contributing to rapidly ensuing pulmonary fibrosis in COVID-19 was identified as a unique feature compared to other causes of pneumonia.
