The method involves the use of DL4-mu beads and lymphopoietic cytokines to induce a sequential differentiation process leading to the development of functional T cells. Single-cell RNA sequencing of human pluripotent stem cell-derived proT cells shows a transcriptional profile similar to early thymocytes. Adoptive transfer of CD34(+)CD7(+) proT cells into immunodeficient mice demonstrates successful thymic engraftment and maturation of peripheral T cells. The DL4-mu bead platform provides a simple and robust tool for studying human T cell development and developing engineered T cell therapies from sustainable sources.
