Global burden of acute lower respiratory infection associated with human metapneumovirus in children under 5 years in 2018: a systematic review and modelling study

Xin Wang et al.

Summary: This study estimated the global burden of human metapneumovirus-associated acute lower respiratory infections in children under 5 years old. In 2018, there were an estimated 14.2 million human metapneumovirus-associated ALRI cases, 643,000 hospital admissions, and 7,700 in-hospital deaths among children younger than 5 years globally. Our findings underscore the importance of intervention strategies to improve outcomes for young infants in low and middle-income countries.

Structure, Immunogenicity, and Conformation-Dependent Receptor Binding of the Postfusion Human Metapneumovirus F Protein

Jiachen Huang et al.

Summary: This study revealed the crystal structure of the hMPV F protein in the postfusion conformation from genotype B, highlighting structural differences between genotypes and receptor binding properties. Vaccine development experiments showed that immunization with postfusion hMPV B2 F protein generated neutralizing antibodies in BALB/c mice, providing protection against hMPV challenge.

Interprotomer disulfide-stabilized variants of the human metapneumovirus fusion glycoprotein induce high titer-neutralizing responses

Guillaume B. E. Stewart-Jones et al.

Summary: The impact of stabilizing disulfides in HMPV F trimers on eliciting neutralizing responses was investigated in mouse and macaque models. Stabilized prefusion and postfusion F trimers induced significantly higher neutralizing responses than non-stabilized F trimers in mice, while in macaques, the impact was more moderate but still significantly higher than average human titers. The absorption-based analyses of macaque responses suggested that IP-DS stabilization not only altered the immunogenicity of epitopes but also their antigenicity.
