Unified classification of mouse retinal ganglion cells using function, morphology, and gene expression

Jillian Goetz et al.

Summary: This study presents a unified classification method for mouse retinal ganglion cells (RGCs) based on visually evoked responses. A total of 1,859 RGCs were classified into 42 types, and morphological and transcriptomic data were used to validate and align the functional classification. An online database was also created, allowing users to browse, download, and classify RGCs using a machine learning algorithm. This work provides a valuable resource for studying RGCs and their circuits in the retina and brain, and establishes a framework for future efforts in neuronal classification and open data distribution.

Patch-seq: Past, Present, and Future

Marcela Lipovsek et al.

Summary: Patch-seq is a revolutionary technique in neuroscience that integrates morphology, physiology, and gene expression data to study neuronal biology comprehensively. It enables targeted study of specific neuronal populations, compilation of multimodal cell type atlases, and investigation of the molecular basis of morphologic and functional diversity. This technique has the potential for further technical development and implementation in various research areas to directly link gene expression to brain function.

Transcriptional and morphological profiling of parvalbumin interneuron subpopulations in the mouse hippocampus

Lin Que et al.

Summary: The study investigated transcriptomic states of morphologically identified parvalbumin interneurons (PV-INs) using single-cell RNA-seq, revealing high transcriptomic similarity among PV-INs with only few genes showing divergent expression. Additionally, PV-INs exhibited a uniform synaptic cell adhesion molecule (CAM) profile, suggesting CAM expression in mature cells does not reflect wiring specificity post-development.

Single-cell profiling of myeloid cells in glioblastoma across species and disease stage reveals macrophage competition and specialization

Ana Rita Pombo Antunes et al.

Summary: By utilizing single-cell RNA sequencing and CITE-seq, this study reveals the complexity of the immune landscape in glioblastomas, including conserved populations of dendritic cells and macrophages. The results indicate that microglia-derived tumor-associated macrophages dominate in newly diagnosed tumors, but are replaced by monocyte-derived TAMs in recurrences.

Single-cell atlas of early human brain development highlights heterogeneity of human neuroepithelial cells and early radial glia

Ugomma C. Eze et al.

Summary: Through single-cell RNA sequencing, nine progenitor populations at the earliest stages of human brain development were identified, including a highly prevalent mesenchymal-like population that disappears once neurogenesis begins. Comparison with mouse progenitor populations revealed two clusters enriched in the early stages of human cortical development. Organoid systems displayed low fidelity to neuroepithelial and early radial glia cell types, but improved as neurogenesis progressed.

Summary: The study introduces a weighted-nearest neighbor analysis framework to learn the relative utility of each data type in each cell, enabling integrative analysis of multiple modalities. Applied to a CITE-seq dataset, the method constructs a multimodal reference atlas of the circulating immune system and successfully identifies and validates previously unreported lymphoid subpopulations.

Phenotypic variation of transcriptomic cell types in mouse motor cortex

Federico Scala et al.

Summary: The study combines single-cell transcriptomic, morphological, and electrophysiological characteristics to classify over 1,300 neurons from the mouse motor cortex, revealing extreme diversity in gene expression, morphology, and electrophysiology. While broad families of transcriptomic types exhibit distinct morpho-electric phenotypes, individual transcriptomic types within the same family show a continuum of variability in morphology and electrophysiology without clear boundaries between them, indicating that neuronal types in the neocortex do not always form discrete entities.
