Molecular Mechanisms of DUBs Regulation in Signaling and Disease

Ying Li et al.

Summary: This review focuses on recent advances in the molecular mechanisms and specificities of DUB enzymes, highlighting examples where the regulatory mechanisms for DUBs have been understood in depth at the molecular level through structural biology. It also discusses strategies used by different DUB families to provide specificity in cleaving particular ubiquitin linkages, as well as recent progress in the development of drug compounds targeting DUB proteases and their correlation to various pathologies such as cancer and neurodegenerative diseases.

Advances in the Development Ubiquitin-Specific Peptidase (USP) Inhibitors

Shiyao Chen et al.

Summary: Ubiquitination and deubiquitination are reversible protein post-translational modification processes that regulate protein degradation under physiological conditions. Imbalance in this regulatory system can lead to diseases like cancer and inflammation. USPs, as main members of the DUBs family, play crucial roles in various molecular signaling pathways and have potential as therapeutic targets. Research focuses on the roles of USPs in different signaling pathways and the development of USP inhibitors.

USP22 promotes melanoma and BRAF inhibitor resistance via YAP stabilization

Ying Wei et al.

Summary: This study identified ubiquitin-specific peptidase 22 (USP22) as a deubiquitinase that controls the abundance and biological functions of Yes-associated protein (YAP) in melanoma. Depletion of USP22 decreases YAP expression, leading to suppressed cell proliferation and tumorigenesis, while overexpression of USP22 confers vemurafenib resistance in a YAP-dependent manner. Overall, the USP22/YAP axis plays an important role in melanoma and BRAF inhibitor resistance, providing a potential target for melanoma treatment.

Pharmacological inhibition of USP7 suppresses growth and metastasis of melanoma cells in vitro and in vivo

Minmin Xiang et al.

Summary: The study revealed that ubiquitin-specific protease-7 is overexpressed in melanoma, acting as an oncogene that promotes melanoma cell proliferation and metastasis. Its inhibitor P22077 effectively inhibits melanoma cell proliferation and induces apoptosis. Additionally, inhibition of ubiquitin-specific protease-7 suppresses melanoma tumor growth and reduces migration and invasion of melanoma cells.

Time-resolved in vivo ubiquitinome profiling by DIA-MS reveals USP7 targets on a proteome-wide scale

Martin Steger et al.

Summary: By combining improved sample preparation, data-independent acquisition mass spectrometry and deep learning, the researchers developed a robust and precise workflow for quantitative ubiquitinome profiling, enabling the characterization of USP7 deubiquitinase targets and inhibitor effects. Their method not only improves system-level understanding of ubiquitin signaling but also increases the identification numbers of ubiquitinated peptides, while dissecting the scope of USP7 action through high temporal resolution analysis.
