Multifunctional hydrogel enhances bone regeneration through sustained release of Stromal Cell-Derived Factor-1? and exosomes

Lang Chen et al.

Summary: Researchers developed an injectable, self-healing, adhesive hydrogel that combines Stromal Cell-Derived Factor-1 alpha (SDF-1 alpha) and M2 macrophage derived exosomes (M2D-Exos). From hurdle to springboard: The macrophage as target in biomaterial-based bone regeneration strategies

Yang-Hee Kim et al.

Summary: In the past decade, there has been an increasing recognition of the role of the innate immune response in mediating repair and biomaterial directed tissue regeneration. The macrophage is now seen as a key player in orchestrating the resolution of inflammation and initiating the process of remodeling and repair. In the context of bone, macrophages have been found to have an essential coordinating role in supporting bone repair and facilitating biomaterial driven osteogenesis. This review examines the evidence for the role of macrophages in bone regeneration and repair, and discusses recent approaches that target macrophage modulation to accelerate and enhance bone tissue regeneration.

Effects of integrated bioceramic and uniaxial drawing on mechanically-enhanced fibrogenesis for bionic periosteum engineering

Wanqi Zhang et al.

Summary: Periosteum plays a crucial role in the management of large bone defects. However, there is limited research on the utilization of periosteum for bone healing in translational medicine. In this study, a bioinspired periosteum scaffold was engineered with the co-delivery of ionic and geometry cues. The scaffold exhibited mechanical anisotropy similar to natural periosteum and supported cell alignment and extracellular matrix deposition. This research provides new insights into periosteum engineering and its potential clinical applications in bone defect repair.

Impact of Digestive Inflammatory Environment and Genipin Crosslinking on Immunomodulatory Capacity of Injectable Musculoskeletal Tissue Scaffold

Colin Shortridge et al.

Summary: The study investigated the effects of the digestive inflammatory environment on scaffold degradation, prevention of premature degradation using genipin crosslinking, and how genipin crosslinking affects IL-4 release and differentiation of naive macrophages. The results showed that the digestive inflammatory environment led to increased protein release from the scaffold, which was improved through genipin crosslinking. The optimal genipin concentration improved IL-4 release, cell viability, mechanical strength, scaffold porosity, and immunomodulation studies, but also decreased the immunomodulatory capacity of the scaffold.
