Hallmarks of Cancer: New Dimensions

Douglas Hanahan

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

Clusters of apoptotic signaling molecule-enriched rafts, CASMERs: membrane platforms for protein assembly in Fas/CD95 signaling and targets in cancer therapy

Faustino Mollinedo et al.

Summary: Mammalian cells have the ability to initiate cell death through death receptors. Cholesterol-rich lipid rafts play a crucial role in the aggregation of Fas/CD95 and the formation of death-inducing signaling complexes (DISC). These raft structures compartmentalize apoptotic signaling machinery and facilitate efficient signal transmission.

Cancer stem cells in TNBC

Zhan Hua et al.

Summary: Triple-negative breast cancer (TNBC) is a type of breast cancer that lacks effective therapeutic targets and has a poorer prognosis compared to other types of breast cancer. TNBC is highly heterogeneous and can include rare cancer stem cells (CSCs) that contribute to treatment resistance. Single cell analysis technologies offer insights into the role of CSCs in TNBC and their potential as therapeutic targets.

Crosstalk between Lipid Rafts and Aging: New Frontiers for Delaying Aging

Shuo Zhang et al.

Summary: With the global population rapidly aging, delaying aging has become a hot research topic. Lipid rafts (LRs), which are microdomains in the plasma membrane containing sphingolipids and cholesterol, have been found to have an interesting interplay with aging. The components of LRs are altered with aging, and LRs strongly influence the aging process. Therefore, targeting LRs is a promising strategy for delaying aging.

Differential glycosylation of collagen modulates lung cancer stem cell subsets through β1 integrin-mediated interactions

Cecilia Gardelli et al.

Summary: The study demonstrates that glycosylation of collagen can promote expansion and generation of cancer stem cells in lung cancer, leading to increased tumorigenicity and metastatic potential. Interference with this glycosylation-mediated crosstalk may offer a novel therapeutic strategy for treating lung cancer.

Induction of Apoptosis in Human Pancreatic Cancer Stem Cells by the Endoplasmic Reticulum-Targeted Alkylphospholipid Analog Edelfosine and Potentiation by Autophagy Inhibition

Consuelo Gajate et al.

Summary: Pancreatic cancer, with its poor prognosis and high mortality rate, remains an incurable malignancy with ineffective current therapies. The study found that the alkylphospholipid analog edelfosine targets pancreatic cancer stem cells, inducing endoplasmic reticulum stress and apoptosis. The combination of endoplasmic reticulum targeting and autophagy inhibitors could offer a new approach in pancreatic cancer treatment involving cancer stem cells.

Lipid raft-disrupting miltefosine preferentially induces the death of colorectal cancer stem-like cells

So-Yeon Park et al.

Summary: This study highlights the important role of lipid rafts (LRs) in colorectal cancer, where an LR-disrupting alkylphospholipid drug can suppress CSC populations and their self-renewal activities. By inhibiting the LR/CHEK1 axis, the drug forces CSCs to enter inappropriate mitosis, leading to catastrophic cell death.

The metabolic flexibility of quiescent CSC: implications for chemotherapy resistance

Kangchen Chen et al.

Summary: Stem cells, including in adults and cancer, exhibit quiescence which can aid cancer stem cells in escaping chemotherapy. The tumor microenvironment provides signals to maintain stem cells in a quiescent state, making targeting TME components a potential treatment for chemotherapy resistance. Modulating metabolic pathways also shows promise in targeting stem cells and TME to limit resistance to chemotherapy.
