Summary: In this study, a hypoxia-induced lncRNA, CBSLR, was discovered to protect gastric cancer cells from ferroptosis, leading to chemoresistance. The mechanism involves the formation of CBSLR/YTHDF2/CBS signaling axis that destabilizes CBS mRNA and promotes ACSL4 degradation, resulting in decreased ferroptosis.

Summary: The RNA binding motif protein 15 (RBM15) has been found to be mutated or abnormally up-regulated in various cancers, and its high expression is associated with poor prognosis. In pancreatic adenocarcinoma (PAAD), RBM15 overexpression is correlated with patients' overall survival, disease-free interval, progression-free interval, and disease-specific survival. RBM15 expression is also positively correlated with immune infiltrating cells and immune checkpoint markers in certain cancer types. Knockdown of RBM15 inhibits the proliferation of pancreatic cancer cells. Pan-cancer analysis suggests that RBM15 may serve as a prognostic biomarker and immunotherapeutic target for PAAD.

Summary: This study reveals that activated JNK signaling is associated with increased expression of METTL3 in bladder cancer. JNK signaling regulates METTL3 expression and global RNA m(6)A levels. Additionally, m(6)A modification is enriched in the 3'-UTR of PD-L1 mRNA, and it is recognized by the m(6)A reader protein IGF2BP1 to mediate the stability and expression levels of PD-L1.

Summary: Habitual areca nut chewing is associated with the high prevalence of oral squamous cell carcinoma (OSCC) in South Asia. This study reveals that chronic arecoline exposure induces upregulation of PD-L1, which confers superior cell proliferation, migration, and resistance to T-cell killing to OSCC cells. Targeting PD-L1 might be a potential therapeutic strategy for treating PD-L1-positive OSCC patients, especially those with habitual areca nut chewing.

Summary: Copper is an essential cofactor for all organisms, but excessive copper can lead to cell death. We found that copper-dependent cell death mechanism is different from known mechanisms, and it is related to mitochondrial respiration. Copper directly binds to lipoylated components of the tricarboxylic acid cycle, causing protein aggregation and loss of iron-sulfur cluster proteins, ultimately leading to cell death.

Summary: This study revealed that the expression of six transmembrane epithelial antigen of the prostate 2 (STEAP2) is downregulated in papillary thyroid cancer (PTC) tissues. Low STEAP2 expression is associated with aggressive clinicopathological characteristics and poor prognosis in PTC patients. The METTL3-mediated STEAP2 N6-methyladenosine (m6A) modification plays a critical tumor-suppressive role in PTC progression. The METTL3-STEAP2 axis may serve as a potential therapeutic molecular target against PTC.
