Updating the Paradigm: Redox Partner Binding and Conformational Dynamics in Cytochromes P450

Summary: This Account summarizes recent findings on the role of redox partner binding, allostery, and conformational dynamics in cytochrome P450 proton coupled electron transfer. The research reveals the heterolytic cleavage of the O-O bond in nearly all P450s, leaving behind a heme iron-linked O atom as the oxidant. Experimentation has shown significant conformational changes and multiple proline isomerizations in P450cam and other bacterial P450s.

Concerning P450 Evolution: Structural Analyses Support Bacterial Origin of Sterol 14α-Demethylases

Summary: Sterol biosynthesis in Methylococcus capsulatus involves a simplified pathway with the key step of sterol 14 alpha-demethylation catalyzed by CYP51. The structure of CYP51fx from M. capsulatus is the first of a bacterial sterol biosynthetic enzyme, suggesting a common evolutionary origin of eukaryotic and bacterial CYP51s. Bioinformatic analysis reveals the presence of CYP51 genes in over 1,000 bacteria, supporting the idea that early eukaryotes may have recruited CYP51 from bacterial sources.
