Detection of germline variants in Brazilian breast cancer patients using multigene panel testing

Rodrigo Santa Cruz Guindalini et al.

Summary: Genetic diversity of germline variants in breast cancer predisposition genes was explored in a study of 1663 Brazilian patients. Pathogenic/likely pathogenic variants were found in 20.1% of participants, with TP53, BRCA1, and BRCA2 being the most commonly mutated genes. The TP53 R337H variant was strongly associated with breast cancer risk, while MUTYH variants showed no association. About 46.1% of patients had variants of uncertain significance (VUS).

Germline variants of Brazilian women with breast cancer and detection of a novel pathogenic ATM deletion in early-onset breast cancer

Gabriel Bandeira et al.

Summary: This study evaluated the frequency of pathogenic/likely pathogenic germline variants in 105 Brazilian women with breast and/or ovarian cancer, identifying P/LP variants in 21% of patients and VUS variants in 14.3% of patients. A novel pathogenic ATM intragenic deletion was also discovered, along with a higher frequency of a BRCA1 pathogenic variant compared to other studies. These findings help characterize the genetic background of breast cancer predisposition in the Brazilian population.

A Population-Based Study of Genes Previously Implicated in Breast Cancer

Chunling Hu et al.

Summary: This study provides population-based estimates of the prevalence and risk of breast cancer associated with pathogenic variants in known cancer-predisposition genes. The results highlight the varying levels of risk associated with different genes, such as high risk for BRCA1 and BRCA2 and moderate risk for PALB2, as well as associations with specific subtypes of breast cancer for genes like RAD51C and RAD51D.

Leila Dorling et al.

Summary: This study sequenced samples from 60,466 women with breast cancer and 53,461 controls using a panel of 34 putative susceptibility genes, identifying associations between protein-truncating variants in certain genes and breast cancer risk. The results provide important information for genetic counseling and define genes that are most clinically useful for predicting breast cancer risk.

Germline molecular data in hereditary breast cancer in Brazil: Lessons from a large single-center analysis

Summary: This study aimed to determine the prevalence of germline pathogenic variants in Brazilian breast cancer patients. The results showed a PV detection rate of 20.5% among the patients, with associations found between PV detection and various high-risk factors for breast cancer. Some patients carried PVs in high-penetrance breast cancer genes, suggesting that multigene panel testing may serve as an effective first-tier diagnostic approach.

Personalized Risk Assessment for Prevention and Early Detection of Breast Cancer: Integration and Implementation (PERSPECTIVE I&I)

Jennifer D. Brooks et al.

Summary: Early detection of breast cancer through screening reduces mortality, but potential harms must be considered. Personalized risk assessment may improve screening programs, and the PERSPECTIVE I&I project aims to identify novel genes, enhance risk prediction tools, develop a socio-ethical framework, and conduct economic analysis for risk-based screening implementation. The goal is to empower women with informed decisions about screening and prevention.
