Real-time identification of two substrate-binding intermediates for the light-driven sodium pump rhodopsin

Summary: Membrane transport proteins undergo critical conformational changes during substrate uptake and release, with at least two substrate-binding intermediates appearing during each transport cycle. The real-time identification of these intermediates for the light-driven Na+-pump rhodopsin revealed independent mechanisms for Na+ uptake and release. The irreversible switch in accessibility observed during the transition from O1 to O2 intermediates may represent a key process governing unidirectional Na+ transport.

These rhodopsins are involved in a variety of light-driven energy and sensory transduction processes.

Summary: Many H+-pump rhodopsins conserve H+ donor residues in cytoplasmic (CP) half channels to quickly transport H+ from the CP medium to Schiff bases. Different types of pumps, such as carboxyl and Lys-type, have optimized mechanisms for their respective H+-transfer reactions. Experimental results show significant differences between archaeal and eubacterial H+ pumps in terms of hydration requirements and donor residue motions during H+-transfer reactions.
