Dopamine promotes the neurodegenerative potential of β-synuclein

Anupam Raina et al.

Summary: The study suggests that dopamine can enhance the neurodegenerative potential of beta-Syn, which may play a significant pathophysiological role in the degeneration of nigral dopaminergic neurons in Parkinson's disease.

Reverse engineering Lewy bodies: how far have we come and how far can we go?

Mohamed Bilal Fares et al.

Summary: Lewy bodies (LBs) are important pathological features of Parkinson disease and other neurodegenerative diseases, with increasing evidence suggesting that the aggregation of alpha-synuclein plays a central role in LB formation. However, little is known about the mechanisms underlying LB formation, their biochemical and ultrastructural properties, evolution and spreading with disease progression, and their role in neurodegeneration.

Comparison of induced neurons reveals slower structural and functional maturation in humans than in apes

Maria Schoernig et al.

Summary: The study showed that chimpanzee and bonobo iNs express genes related to dendrite and synapse development earlier than human iNs, leading to faster development in the former two species. However, all three species eventually reach the same level of structural complexity.

Neuroprotective effects of garlic extract on dopaminergic neurons of substantia nigra in a rat model of Parkinson's disease: motor and non-motor outcomes

Maryam Bigham et al.

Summary: Parkinson's disease is a common and severe neurodegenerative disorder that involves a selective loss of dopaminergic neurons. Studies have shown that garlic extract can protect cells from oxidative stress and inflammation, and improve movement disorders and behavioral deficits in animal models of PD.

Administration of quercetin improves mitochondria quality control and protects the neurons in 6-OHDA-lesioned Parkinson's disease models

Wen-Wen Wang et al.

Summary: Studies show that quercetin can improve mitochondrial quality control, reduce oxidative stress, increase levels of mitophagy markers PINK1 and Parkin, and decrease alpha-synuclein expression in 6-OHDA-treated cells. In vivo, quercetin also helps alleviate PD-like motor behaviors, decrease neuronal death, reduce mitochondrial damage, and control alpha-synuclein accumulation in PD rats. The neuroprotective effect of quercetin is compromised when either Pink1 or Parkin is knocked down.

The LRRK2 G2019S mutation alters astrocyte-to-neuron communication via extracellular vesicles and induces neuron atrophy in a human iPSC-derived model of Parkinson's disease

Aurelie de Rus Jacquet et al.

Summary: This study focused on the effects of a Parkinson's disease-related mutation on astrocytes and neurons, revealing alterations in extracellular vesicle biogenesis and abnormal accumulation of PD-related proteins. The findings suggest that dysfunctional astrocyte-to-neuron communication via altered EV biological properties may participate in the progression of Parkinson's disease.

Generation of two gene edited iPSC-lines carrying a DOX-inducible NGN2 expression cassette with and without GFP in the AAVS1 locus

Benjamin Schmid et al.

Summary: Neurog2 is a gene encoding the neuronal transcription factor NGN2, converting stem cells into functional neurons efficiently. Two iPS cell lines, BIONi010-C-13 and BIONi010-C-15, were generated with DOX-inducible constructs of neurog2, allowing differentiation into neurons when DOX is added.

BANP opens chromatin and activates CpG-island-regulated genes

Ralph S. Grand et al.

Summary: The study reveals that BANP is a critical transcription activator that binds to the CGCG element, controlling essential metabolic genes in pluripotent stem cells and neuronal cells. BANP binding is influenced by DNA methylation, restricting most binding to CGIs and explaining differential binding at CGI promoters in cancer cells. Additionally, BANP's binding to unmethylated motifs leads to chromatin opening and nucleosome phasing.

Stem cell-derived neurons reflect features of protein networks, neuropathology, and cognitive outcome of their aged human donors

Valentina N. Lagomarsino et al.

Summary: This resource includes iPSC lines from 53 individuals, coupled with RNA and proteomic profiling of both iPSC-derived neurons and brain tissue. Analysis of iPSC neurons revealed associations between specific Ab and tau species and cognitive decline, as well as identified proteins and networks associated with AD phenotypes.

SETBP1 accumulation induces P53 inhibition and genotoxic stress in neural progenitors underlying neurodegeneration in Schinzel-Giedion syndrome

Federica Banfi et al.

Summary: Schinzel-Giedion syndrome (SGS) is a fatal developmental syndrome characterized by severe intellectual and physical deficits due, at least in part, to early neurodegeneration. The neuronal death in SGS originates from developmental alterations mainly in safeguarding cell identity and homeostasis.

Carol X-Q Chen et al.

Summary: A multistep workflow has been established to evaluate newly generated iPSCs, testing key benchmarks such as cell growth, genomic stability, pluripotency, and the ability to differentiate into specific cell types. Simple cell growth tests and genomic integrity analysis are introduced to ensure quality control of iPSCs lines.
