Longitudinal analysis reveals high prevalence of Epstein-Barr virus associated with multiple sclerosis

Summary: This study suggests that multiple sclerosis may be caused by Epstein-Barr virus, with no direct association with other viral infections. These findings provide clues to the primary cause of MS.

A comparative study of melatonin and immunomodulatory therapy with interferon beta and glatiramer acetate in a mouse model of multiple sclerosis

E. J. Ramos Gonzalez et al.

Summary: The present study compared the effects of MLT administration with IFN-beta and GA in an EAE rat model. Results showed that IFN-beta and GA had better clinical evaluation results compared to MLT. Further research is needed to evaluate the molecular mechanisms of MLT in EAE and quantify other cytokines in different biological media to determine if MLT can be considered as an anti-inflammatory agent for regulating MS.

Nicotinamide adenine dinucleotide treatment alleviates the symptoms of experimental autoimmune encephalomyelitis by activating autophagy and inhibiting the NLRP3 inflammasome

Xin Wang et al.

Summary: The study showed that treatment with NAD+ alleviated demyelination, nerve injury, and microglial activation in EAE mice by activating autophagy. NAD+ also suppressed the NLRP3 inflammasome, modulated Th1 and Th17 cells, reduced pro-inflammatory factors, and increased anti-inflammatory factors. Conversely, inhibition of autophagy using 3-MA aggravated inflammation and delayed remission from EAE, and abolished the beneficial effects of NAD+.

Tryptophan: A Rheostat of Cancer Immune Escape Mediated by Immunosuppressive Enzymes IDO1 and TDO

Minah Kim et al.

Summary: Blocking tryptophan catabolism mediated by IDO1 and TDO shows promise for sensitising cancer patients to immune checkpoint blockade. Understanding the immune-regulatory function of tryptophan catabolism could lead to optimized therapeutic strategies. Tryptophan may act as a rheostat of kynurenine-mediated immunosuppression by competing for entry into immune T-cells.

Tryptophan-kynurenine metabolism: a link between the gut and brain for depression in inflammatory bowel disease

Li-Ming Chen et al.

Summary: Inflammatory bowel disease (IBD) patients are at a higher risk of depression, which can negatively impact disease prognosis and quality of life. The key metabolic enzyme indoleamine 2,3-dioxygenase (IDO) in the tryptophan-kynurenine metabolic pathway is activated in the inflammatory environment, leading to potentially neurologically active metabolites associated with inflammation-induced depression.

The kynurenine pathway in chronic diseases: a compensatory mechanism or a driving force?

Niklas Joisten et al.

Summary: The dysregulation of the kynurenine pathway in tryptophan metabolism is associated with inflammation-driven pathologies, making it a potential therapeutic target for diseases such as cancer and brain disorders. It is important to consider both compensatory mechanisms and bioenergetic roles of KP metabolites in designing effective therapeutic approaches to attenuate disease progression.

A Novel NAD Signaling Mechanism in Axon Degeneration and its Relationship to Innate Immunity

Eleanor L. Hopkins et al.

Summary: Axon degeneration is a pathological feature of many neurodegenerative diseases, regulated by the key proteins NMNAT2 and SARM1. The interaction between NMNAT2 and SARM1 plays a crucial role in maintaining axon integrity, and this pathway can be activated in various conditions, leading to axon pathology.
