Synthesis of chiral sulfinate esters by asymmetric condensation

Summary: Chiral sulfur functional groups are often neglected in drug discovery, but they have the potential to expand the chemical space. In this study, we report a straightforward access to enantioenriched sulfinate esters using pentanidium as an organocatalyst. These sulfinate esters can be transformed into diverse chiral sulfur pharmacophores, demonstrating the viability of this approach towards sulfur stereogenic centers through late-stage diversification.

Chiral Arene Ligand as Stereocontroller for Asymmetric C-H Activation

Summary: In this study, the combination of an achiral cobalt(III) catalyst and a pseudo-C-2-symmetric H-8-binaphthyl chiral carboxylic acid was utilized for the asymmetric synthesis of benzothiadiazine-1-oxides containing a chiral sulfur center. The optimized protocol achieved high enantioselectivity and allowed for the synthesis of benzothiadiazine-1-oxides with various functional groups.

Asymmetric Ru/Cinchonine Dual Catalysis for the One-Pot Synthesisof Optically Active Phthalides from Benzoic Acids and Acrylates

Summary: In this study, we report an asymmetric Ru/cinchonine dual catalysis for the enantioselective synthesis of C-3 substituted phthalides through tandem C-H activation/Michael addition cascade. The use of readily accessible and less expensive catalysts greatly overcomes the trend of using highly sophisticated catalysts. The developed method allows access to both enantiomers of the product and streamlines the synthesis of optically active phthalides.

Ir(III)-Catalyzed Asymmetric C-H Activation/Annulation of Sulfoximines Assisted by the Hydrogen-Bonding Interaction

Summary: The Ir(III)-catalyzed asymmetric C-H activation enabled by noncovalent interactions allows for the preparation of a broad range of sulfur-stereogenic sulfoximines with high yields and excellent enantioselectivities. Detailed DFT calculations and experimental studies have shown that the N-H···O hydrogen bonding interaction between sulfoximine and the chiral carboxylic acid ligand is crucial for the high enantiocontrol. Furthermore, chiral iridacycle intermediates have been isolated, characterized, and subjected to stoichiometric reactions, indicating that the C-H cleavage step is the rate- and enantio-determining step.

Efficient Synthesis of Sulfur-Stereogenic Sulfoximines via Ru(II)-Catalyzed Enantioselective C-H Functionalization Enabled by Chiral Carboxylic Acid

Summary: An undeveloped Ru(II)-catalyzed enantioselective C-H functionalization involving an enantiodetermining C-H cleavage step has been described in this study. Using a novel class of chiral binaphthyl monocarboxylic acids as chiral ligands, a broad range of sulfur-stereogenic sulfoximines were synthesized in high yields with excellent enantioselectivities (up to 99% yield and 99% ee) via desymmetrization, kinetic resolution, and parallel kinetic resolution. Furthermore, the resolution products can be easily transformed into chiral sulfoxides and key intermediates for kinase inhibitors.

Enantioselective Ruthenium-Catalyzed C-H Alkylations by a Chiral Carboxylic Acid with Attractive Dispersive Interactions

Summary: The asymmetric ruthenium-catalyzed C-H alkylations were achieved using a chiral C2-symmetric carboxylic acid, resulting in chiral tetrahydrocarbazoles and cyclohepta[b]indoles with high enantioselectivity at room temperature. Mechanistic studies revealed a fast C-H ruthenation process, with a rate- and enantio-determining proto-demetalation step. The asymmetric induction was governed by weak attractive secondary dispersion interactions, as shown in NCI analysis of the key transition states.
