A Founder Mutation in EHD1 Presents with Tubular Proteinuria and Deafness

Summary: This study identified a previously unrecognized autosomal recessive disorder characterized by sensorineural deafness and tubular proteinuria caused by a homozygous missense variant of EHD1. Functional studies in patients and animal models revealed that the mutation impairs receptor-mediated endocytosis in proximal tubules, leading to impaired reabsorption of filtered proteins.

Summary: Proteinuria is a well-established marker and predictor of kidney disease, and the study found that segments 1 and 2 of the proximal tubule are responsible for clearing protein filtrate in normal physiological conditions. However, under proteinuric stress, segment 3 can also participate in the reabsorption process. The endolysosomal system was most prominent in segments 1 and 2, with varied depth of ligand uptake among nephrons.

A Comprehensive Map of mRNAs and Their Isoforms across All 14 Renal Tubule Segments of Mouse

Summary: This study used a full-length, small-sample RNA-seq protocol to profile transcriptomes for all 14 renal tubule segments microdissected from mouse kidneys, identifying over 34,000 transcripts and 3709 segment-specific transcripts. Gene-expression patterns of solute carriers, transcription factors, and G protein-coupled receptors along the renal tubule were found to be important for segment-specific function.
