A structure of human Scap bound to Insig-2 suggests how their interaction is regulated by sterols

Renhong Yan et al.

Summary: The sterol regulatory element-binding protein (SREBP) pathway is controlled by membrane-embedded sterol sensors Scap, Insig-1 and Insig-2, with 25-hydroxycholesterol (25HC) serving as a master switch. Cryo-electron microscopy analysis revealed the structure of the human Scap and Insig-2 complex in the presence of 25HC, showing the critical role of 25HC binding and Insig association in regulating the pathway.

The eukaryotic MEP-pathway genes are evolutionarily conserved and originated from Chlaymidia and cyanobacteria

Liping Zeng et al.

Summary: This study analyzes the evolutionary history of MEP-pathway genes, showing their polyphyletic origin and restricted conservation. It also highlights the indispensable function of individual enzymes in the MEP pathway beyond isoprenoid production, supported by their high expression levels and distinct phenotypic variations in plants with reduced expression levels.

Harzianic Acid from Trichoderma afroharzianum Is a Natural Product Inhibitor of Acetohydroxyacid Synthase

Linan Xie et al.

Summary: Acetohydroxyacid synthase (AHAS) is a crucial enzyme in the branched-chain amino acid biosynthetic pathway and harzianic acid (HA) produced by the biocontrol fungus has been identified as a natural product inhibitor of AHAS. HA selectively inhibits fungal AHAS, including resistant mutants, through a competitive inhibition mechanism different from known commercial inhibitors. The biosynthesis of HA by the fungus in the rhizosphere is proposed as an important role based on the study data.
