Host protease activity classifies pneumonia etiology

Melodi Anahtar et al.

Summary: This study developed a panel of nanosensors to detect the activity of pulmonary host proteases and differentiate between bacterial and viral pneumonia based on urinary readouts. Machine learning algorithms were used to train diagnostic classifiers for accurate diagnosis of infected mice.

Protease Activity Analysis: A Toolkit for Analyzing Enzyme Activity Data

Ava P. Soleimany et al.

Summary: Analyzing protease and substrate activity is crucial for understanding their biological functions and developing new diagnostic and therapeutic methods. However, there is a lack of computational tools for automating the analysis of in vitro and in vivo protease assays, leading to a lack of standardization in the field. To address this, we present Protease Activity Analysis (PAA), a toolkit that provides a modular framework for preprocessing, visualization, machine learning analysis, and querying of protease activity data sets.

Microenvironment-triggered multimodal precision diagnostics

Liangliang Hao et al.

Summary: Multimodal nanosensors have been developed to target and respond to hallmarks in the tumor microenvironment and provide both a non-invasive urinary monitoring tool and an on-demand positron emission tomography imaging agent to localize tumor metastasis and assess response to therapy. Through tailored target specificities, this modular platform has the capacity to be engineered as a pan-cancer test that may guide treatment decisions for numerous tumor types.

Activatable Zymography Probes Enable In Situ Localization of Protease Dysregulation in Cancer

Ava P. Soleimany et al.

Summary: Recent advancements in conditionally activated diagnostics and therapeutics for cancer have been enabled by the development of novel in situ probes that can detect protease activity within tissue microenvironments. These activatable zymography probes (AZPs) have the potential to provide new insights into protease dysregulation in cancer and accelerate the development of tailored cancer diagnostics and therapeutics.
